摘 要:断奶腹泻是仔猪断奶后常见且危害性较大的应激性疾病。为探究仔猪肠道黏蛋白与断奶腹泻的关系,揭示MUC12在抵抗断奶腹泻中的作用,试验分别将35日龄民猪仔猪48头、长白猪仔猪60头分为断奶腹泻组、断奶健康组、未断奶组(对照组),屠宰后观察仔猪肠道杯状细胞的分布与形态,并对仔猪各肠段杯状细胞进行着色度统计,利用qRT-PCR方法检测MUC12 mRNA在十二指肠、空肠、回肠、盲肠和结肠的相对表达量等。结果表明:杯状细胞分布于各肠段,被染成紫红色,顶部膨大,底部窄小,轮廓清晰;未断奶组民猪仔猪十二指肠和空肠(P<0.01)及断奶健康组仔猪十二指肠(P<0.05)、空肠和回肠(P<0.01)的细胞着色度高于长白猪仔猪;MUC12 mRNA在所有民猪与长白猪仔猪盲肠和结肠中均表达明显,但在十二指肠、空肠和回肠中仅微量表达,其中未断奶组和断奶健康组的民猪仔猪在空肠、回肠和结肠的MUC12 mRNA相对表达量均高于长白猪仔猪。说明民猪肠道分泌的黏蛋白含量高于长白猪,这是民猪断奶腹泻程度低于长白猪的重要生理基础。Post-weaning diarrhea is a common and harmful stress disease in piglets.In order to investigate the relationship between intestinal mucin 12 and post-weaning diarrhea in piglets and to reveal the role of MUC12 in resisting post-weaning diarrhea,forty-eight 35-day-old Min piglets and 60 landrace piglets were divided into post-weaning diarrhea group,healthy weaning group and non-weaning group(control group).The distribution and morphology of goblet cells in the intestine of piglets were investigated after slaughter,and the staining statistics of goblet cells in the duodenum,jejunum,ileum,cecum and colon were carried out.The relative expression of MUC12 mRNA in 5 intestinal segments was detected by qRT-PCR.The results showed that the goblet cells were stained purple and distributed in each intestinal segment and shown to be with the enlarged top and narrow and clear bottom.The coloredness of goblet cells of the duodenum and jejunum(P<0.01)of Min piglets in unweaned group and of the duodenum(P<0.05),jejunum and ileum(P<0.01)of Min piglets in weaned healthy group were higher than that in landrace piglets.MUC12 mRNA expression was found significant in cecum and colons,but slight in duodenum,jejunum and ileum in all pigs of groups,among which,MUC12 mRNA expression in jejunum,ileum and colon of Min piglets in unweaned group and in weaned healthy group was higher than that of Landrace piglets.It can be concluded that the higher intestinal expression of MUC12 in Min piglets than that of Landrace could be an important physiological factor for the lower incidence of post-weaning diarrhea in Min piglets than in landrace piglets.
