气道滴注PM2.5混悬液并灌胃给予橄榄苦苷的去势雌性大鼠股骨骨密度及骨矿物盐含量变化

摘  要:目的观察气道滴注PM2.5混悬液并灌胃给予橄榄苦苷的去势雌性大鼠股骨骨密度及骨矿物盐含量变化。方法30只8周龄SPF级雌性SD大鼠去势后,随机分为观察组和对照组,观察组又分为观察组1、观察组2,每组10只。观察组1气道滴注PM2.5混悬液16.5μg并50 mg/kg橄榄苦苷溶液灌胃、每3天1次(第1次气道滴注时为第1天)共150 d,观察组2灌胃橄榄苦苷溶液为100 mg/kg、其余同观察组1,对照组气道滴注PM2.5混悬液16.5μg并等量生理盐水灌胃。结果与对照组比较,观察组大鼠左股骨干骺端及左股骨中段骨密度高(P均<0.01)。与左股骨中段比较,对照组、观察组大鼠左股骨干骺端骨密度高(P均<0.01);与观察组1比较,观察组2大鼠左股骨干骺端骨密度高(P<0.01)。与对照组比较,观察组大鼠左股骨中段与左股骨干骺端骨密度差值高(P均<0.01)。结论相比于气道滴注PM2.5混悬液大鼠,气道滴注PM2.5混悬液并灌胃给予橄榄苦苷的去势雌性左股骨干骺端、左股骨中段骨密度高,且100 mg/kg橄榄苦苷灌胃处理的效果好。Objective To observe the changes of bone mineral density(BMD)and bone mineral salt content in femur of ovariectomized female rats who were given PM2.5 suspension by intratracheal instillation and oleuropicrin by gavage.Methods Thirty 8-week-old SPF female SD rats were randomly divided into the observation group and control group after castration.The rats in the observation group were further divided into the observation group 1 and observation group 2,with 10 rats in each group.In the observation group 1,airway instillation of PM2.5 suspension 16.5μg and intragastric administration of 100 mg/kg oleuropein solution were given once every 3 days(the time of the first airway instillation was taken as the first day)for 150 days.In the observation group 2,50 mg/kg oleuropicroside solution was administered by gavage,and the rest treatment was the same as that of the observation group 1.In the control group,airway instillation of 16.5μg PM2.5 suspension and the intragastric administration of same amount of normal saline were given.Results Compared with the control group,the bone mineral density of the left femoral metaphysis and the middle segment of the left femur in the observation group were higher(both P<0.01).Compared with the middle segment of the left femur,the bone mineral density(BMD)of the left femoral metaphysis in the control group and the observation group was higher(P<0.01).Compared with the observation group 1,the BMD of left femoral metaphysis in the observation group 2 was higher(P<0.01).Compared with the control group,the BMD of the middle segment and the epiphyseal end of the left femoral shaft in the observation group was higher(P<0.01).Conclusion Compared with rats receiving PM2.5 suspension via airway instillation,ovariectomized females receiving PM2.5 suspension via airway instillation and oral administration of oleuropicrin have higher BMD in the epiphysis and middle segment of left femur,and the effect of oral administration of 100 mg/kg oleuropicrin is better.
