摘 要:以川西獐牙菜作为试验材料,利用PCR技术获得川西獐牙菜MYB基因的全长c DNA序列,运用NCBI、CBS和Ex PASy网站中的信息分析工具,结合Mega 7.0等生物信息分析软件,分析预测MYB基因编码蛋白结构的特征和功能。结果显示,生物信息学预测MYB蛋白的分子式为CHNOS,MYB蛋白不含跨膜区,无信号肽,具有一个保守结构域,其开放阅读框全长1158 bp,编码385个氨基酸,其中丝氨酸的含量最高;理论等电点为6.02,属于亲水性蛋白;系统发育分析显示该蛋白与三龙胆R2R3转录因子在同一个分支上,表示两者间亲缘性较高。本试验结果为进一步研究该基因的功能及利用基因工程手段提高川西獐牙菜中环烯醚萜类化合物产量提供了理论依据。Based on the MYB gene transcript sequence in the transcriptome data of Swertia mussotii obtained from the research group,the full-length c DNA sequence of the MYB gene of Swertia mussotii is obtained by PCR.Use the information analysis tools in the NCBI,CBS and Ex PASy websites,combined with bioinformatics analysis software such as Mega 7.0,to analyze and predict the characteristics and functions of the protein structure encoded by the MYB gene.The bioinformatics predicted that the molecular formula of MYB gene is CHNOS.The MYB protein contain no transmembrane region,no signal peptide,and has a conserved domain.Its open reading frame is 1 158 bp in length,encoding 385 amino acids,and the content of serine is the highest.The theoretical isoelectric point is 6.02,which is a hydrophilic protein.Phylogenetic analysis shows that the gene is on the same branch as the R2R3 transcription factor of Gentiana ternifolia,indicating that the relationship between the two is high.The results of this experiment provide a theoretical basis for further research on the function of this gene and the use of genetic engineering methods to improve the yield of iridoids in Swertia mussotii.
