高温水氨解快速提取人发角蛋白

摘  要:为了获得提取时间短、蛋白质破坏小的人发角蛋白,采用高温水氨解对人发角蛋白进行了快速提取,并用聚丙烯酰胺凝胶电泳(SDS-PAGE)、傅里叶红外光谱(FT-IR)和X-射线衍射(XRD)对所得样品进行了分析。结果表明,185℃、8 mL氨水、降解30 min,可获得分子量分布在25 kDa以下、35~48 kDa、75~135 kDa的人发角蛋白冻干粉末,分子量破坏较小。角蛋白α-螺旋和β-折叠结构减少,晶体化程度降低,具有典型的蛋白质结构,且未引入明显杂质。本研究证实,高温水氨解是一种快速提取人发角蛋白的方法。In order to obtain human hair keratin with short extraction time and little damage, human hair keratin is rapidly extracted by high-temperature water ammonolysis. The obtained samples are analyzed by polyacrylamide gel electrophoretic(SDS-PAGE), fourier transform infrared spectroscopy(FT-IR) and X-ray diffraction(XRD). The results show that human hair keratin freeze-dried powder with molecular distribution below 25 kDa, 35 ~ 48 kDa,75 ~ 135 kDa can be obtained at 185 ℃ with 8 mL ammonia for 30 min degradation. The molecular weight is less destroyed.The α-helix and β-fold structures of extracted keratins are reduced, and the degree of crystallization is reduced. The obtained keratin has a typical protein structure, and no obvious impurities are introduced. The results indicated that high-temperature water ammonolysis is a good method for rapid extraction of human hair keratin.
