摘  要:人乳头瘤病毒(HPV)是一种常见的传染性病原体,目前与HPV感染相关的宫颈、肛门和口咽癌发病率持续增高,已经成为当下全球癌症防治的热点。由于HPV的生命周期对多层上皮宿主细胞分化程序的依赖性以及种属特异性,使得建立一个适合的HPV体外感染模型十分困难。随着三维(3D)细胞培养系统的出现,克服了这一障碍,这一系统完整地再现了上皮细胞的分化,为完成HPV的生命周期提供了一个良好的生存环境,是更好地研究HPV相关癌症的发生发展以及治疗药物的体外模型。本文将从HPV特性的角度分析3D细胞培养在HPV相关癌症研究中的优势,介绍适用的3D细胞培养方式,总结分析3D细胞培养在HPV相关癌症研究中应用及展望,为优化研究HPV相关癌症的3D细胞培养模型提供理论依据。Human papillomavirus(HPV)is a common infectious pathogen,and the incidence of cervical,anal and oropharyngeal cancers associated with HPV infection continues to increase and has become a current global hotspot for cancer prevention and treatment.The dependence of the HPV life cycle on the differentiation program of the multilayer epithelial host cells and its species specificity make it difficult to establish a suitable HPV infection model in vitro.This obstacle has been overcome with the advent of the three-dimensional(3D)cell culture system,a system that completely recapitulates epithelial cell differentiation,provides a good survival environment for completing the HPV life cycle,and is a better in vitro model for studying the development of HPV-associated cancers as well as therapeutic agents.This paper analyzes the advantages of 3D cell culture in HPV-associated cancer research from the perspective of HPV characteristics,introduces applicable 3D cell culture methods,summarizes as well as analyzes the application and outlook of 3D cell culture in HPV-associated cancer research so as to provide a theoretical basis for optimizing 3D cell culture models for studying HPV-associated cancers.
