DC-CIK逆转卵巢癌耐药细胞SKOV3/DDP多药耐药的研究

摘　　要：目的观察体外培养的树突状细胞(DC)联合细胞因子诱导的杀伤细胞(CIK)对卵巢癌耐药株SKOV3/DDP的逆转耐药作用及其对耐药基因MDR1表达量的影响。方法采集健康人外周血单核细胞,用相应细胞因子诱导成DC和CIK;CCK法检测经CIK和DC-CIK作用的SKOV3/DDP细胞对顺铂敏感性的变化;实时荧光定量RT-PCR检测SKOV3组、SKOV3/DDP组、SKOV3/DDP+CIK组、SKOV3/DDP+DC-CIK组的耐药基因MDR1表达量的变化。结果顺铂对经CIK和DC-CIK作用后的SKOV3/DDP细胞的IC50由22.33μg/mL下降到14.31μg/mL、11.89μg/mL,逆转倍数分别为1.56倍和1.96倍;SKOV3/DDP组、SKOV3/DDP+CIK组、SKOV3/DDP+DC-CIK组的MDR1表达量比SKOV3组高(F=23.825,P=0.000),后两组的MDR1表达量较SKOV3/DDP组均下降,SKOV3/DDP+DC-CIK组下降更明显(t=2.316,P=0.047)。结论DC-CIK和CIK均能增加SKOV3/DDP对DDP的敏感性,部分逆转其多药耐药作用,均能下调耐药细胞中的MDR1基因,且DC-CIK作用更明显。Objective To observe the effects of dendritic cells co-cultured with cytokine induced killer cells revesing multidrug resistance of the ovarian cancer resistant cell SKOV3/DDP and the variation of multi-resistant MDR1 gene expression in vitro.MethodsMononuclear cells were isolated from the peripheral blood of healthy individuals and induced into DC and CIK with corresponding cytokine respectively.After CIK and DC-CIK respectively preprocessing,detct the SKOV3/DDP cells sensitivity to cisplatin with CCK kit;detect the MDR1 gene mRNA expression of SKOV3,SKOV3/DDP,SKOV3/DDP+CIK and SKOV3/DDP+DC-CIK by real Time RT-PCR.ResultsAfter CIK and DC-CIK preprocessing,the IC50 of cisplatin to SKOV3/DDP were decreased to 14.31μg/mL、11.89μg/mL from^(2)2.33μg/mL,reverse multiples respectively were 1.56 and 1.96;the expression of MDR1 of SKOV3/DDP,SKOV3/DDP+CIK,SKOV3/DDP+DC-CIK were higher than SKOV3(F=23.825,P=0.000),both MDR1 of SKOV3/DDP+CIK and SKOV3/DDP+DC-CIK expressed lower than SKOV3/DDP,but MDR1 of SKOV3/DDP+DC-CIK cell expressed lowest(t=2.316,P=0.047).ConclusionBoth CIK and DC-CIK can increase the sensitivity of SKOV3/DDP to cisplatin,partly reversing the drug resistance,and down-regulate the MDR1 expression,but the effects of DC-CIK are stronger.
