益肾通络方对雌性小鼠卵子修复精子DNA能力的影响

摘　　要：目的:探讨益肾通络方是否能够改善卵母细胞的DNA修复能力。方法:复制苯并(a)芘(BaP)致雄鼠精子DNA损伤模型,使正常雄鼠(雄性空白对照组)及BaP染毒雄鼠(雄性造模组)分别与灌胃益肾通络方水煎液及灌胃生理盐水的雌鼠合笼,对比各组雌鼠的妊娠结局。结果:造模后,雄性造模组小鼠精子DNA碎片指数(DFI)高于雄性空白对照组(P<0.01)。与空白组(正常雄鼠与正常雌鼠合笼)比较,同BaP染毒雄鼠合笼的正常雌鼠(BaP组)产仔数及活胎数显著降低(P<0.01),死胎率明显增高(P<0.01);与BaP组比较,同BaP染毒雄鼠合笼的灌胃益肾通络方雌鼠(益肾通络方组)产仔数及活胎数显著增加(P<0.05,P<0.01),但两组死胎数比较,差异无统计学意义。结论:益肾通络方提前干预雌鼠可改善其与高精子DNA碎片指数(DFI)雄鼠合笼的妊娠结局。具体机制推测可能与益肾通络方改善卵子的DNA修复能力有关。Objective:To investigate whether Yishen Tongluo Formula can improve the DNA repair ability of oocytes.Methods:The sperm DNA damage model was induced by benzo(a)pyrene(BaP)in male mice.The normal male mice(male normal control group)and BaP infection male mice(male modeling group)were cooped up with female mice administrated with Yishen Tongluo Formula and normal saline,respectively.The pregnancy outcome was compared among the groups.Results:DNA fraction index(DFI)was higher in the male modeling group than that in the male blank control group after modeling(P<0.01).The litter size and live fetus number were significantly decreased(P<0.01),the stillbirth rate was significantly increased in the BaP group(female mice cooped with BaP male mice),when compared with the normal blank group(the normal female mice cooped up with normal male mice)(P<0.01).Compared with the BaP group,the number of births and live births in the Yishen Tongluo Formula group(female mice administrated with Yishen Tongluo Formula and cooped up with BaP infection male mice)were significantly increased(P<0.05,P<0.01),but there was no statistical difference in the number of stillbirths between the two groups.Conclusion:Intervention of Yishen Tongluo Formula in advance can improve the pregnancy outcome of the female mice mating with the male mice with high DFI.The specific mechanism may be related to the improvement of DNA repair ability of ovum by Yishen Tongluo Formula.
