摘 要:目的:探讨定坤丹口服液与定坤丹水蜜丸对雌性去势大鼠模型的性激素调节作用,并对两种剂型的药效进行对比。方法:将雌性SD大鼠随机分为空白组、模型组、阳性对照组(给予补佳乐0.12 mg/kg)、定坤丹水蜜丸组(给药剂量为1.75 g/kg)以及定坤丹口服液高、中、低剂量组(给药剂量分别为10、5、2.5 mL/kg)。除空白组外,其余大鼠采用手术去除双侧卵巢的方法建立去势大鼠模型,术后1周开始灌胃给药,每天灌胃1次,连续灌胃4周。给药期间观察大鼠一般状态;ELISA法检测大鼠血清中卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E_(2))、孕酮(P)、前列腺素E_(2)(PGE_(2))及前列腺素F2α(PGF2α)水平的变化情况;取全血放入促凝管中测定凝血功能;摘取子宫计算脏器系数,并通过HE染色法检测大鼠子宫的病理变化。结果:给药期间,与模型组相比,定坤丹口服液中剂量组大鼠体质量明显增加(P<0.01),与定坤丹水蜜丸组相比差异无统计学意义(P>0.05);子宫指数结果显示,与模型组相比,定坤丹口服液各剂量组、定坤丹水蜜丸组及阳性对照组大鼠子宫指数均较模型组显著升高(P<0.01);凝血功能结果显示,与模型组相比,定坤丹口服液高剂量组大鼠的PT值显著降低(P<0.05),定坤丹口服液高、中剂量组及定坤丹水蜜丸组大鼠的TT值较模型组明显降低(P<0.01,P<0.05),定坤丹口服液与定坤丹水蜜丸组大鼠PT及TT值相比差异无统计学意义(P>0.05);ELISA结果显示,与模型组相比,定坤丹口服液各剂量组、定坤丹水蜜丸组大鼠血清P、E_(2)、PGE_(2)水平均不同程度升高,LH、FSH、PGF2α水平均不同程度降低(P<0.05或P<0.01),其中定坤丹口服液高剂量组大鼠PGE_(2)水平、中剂量组大鼠FSH的水平优于定坤丹水蜜丸组(P<0.01,P<0.05);病理检查结果显示,与模型组相比,各给药组大鼠子宫均有不同程度的萎缩,水蜜丸组及定坤丹口服液高剂量组大鼠子Objective:To investigate the regulatory effects of Dingkundan oral liquid and Dingkundan waterhoneyed pill in ovariectomized rats,and to compare the efficacy differences of the two dosage forms.Methods:Female SD rats were randomly divided into the blank group,the model group,the positive control group(Estradiol Valerate Tablet,0.12 mg/kg),the Dingkundan water-honeyed pill group(1.75 g/kg),and the Dingkundan oral liquid groups of high-dose,medium-dose and low-dose(10 mL/kg,5 mL/kg,2.5 mL/kg).The ovariectomized rat model was established by the method of removing both ovaries.One week after modeling,the rats were given corresponding interventions.General state of the rats was observed,and the serum levels of FSH,LH,E_(2),P,PGE_(2)and PGF2αwere detected by ELISA method.The blood was put into the coagulation tube to measure the coagulation function.The uterus was extracted to calculate the organ coefficient,and the pathological changes of the uterus were detected in the rats by HE staining method.Results:During administration,compared with that in the model group,body mass was increased significantly in the Dingkundan oral liquid group of medium-dose(P<0.01),of which there was no statistical difference between Dingkundan water-honeyed group and the Dingkundan oral liquid group of medium-dose(P>0.05).Compared with that in the model group,the uterine index was significantly increased in the Dingkundan oral liquid groups,Dingkundan waterhoneyed pill group and the positive control group(P<0.01).Coagulation function results showed that compared with those in the model group,PT value was significantly decreased in the Dingkundan oral liquid group of highdose(P<0.05),TT value was significantly decreased in the Dingkundan oral liquid groups of high-dose and medium-dose and in the Dingkundan water-honeyed pill group(P<0.01,P<0.05);there were no statistical differences in PT value and TT value between the Dingkundan oral liquid groups and the Dingkundan waterhoneyed pill group(P>0.05).ELISA results showed that compared with th
