摘 要:目的:建立凤仙透骨草药材高效液相色谱(HPLC)指纹图谱,并探究特征图谱与抗炎活性间的谱效关系。方法:采用HPLC建立凤仙透骨草药材的指纹图谱;利用聚类分析及主成分分析对共有峰进行评价;建立角叉菜胶致小鼠急性炎症模型,考察不同来源凤仙透骨草药材抗炎活性,利用灰色关联度分析和偏最小二乘判别分析(PLS-DA)探究谱效关系。结果:建立了凤仙透骨草药材的指纹图谱,确定15个共有峰,相似度均在0.90以上,并确认了阿魏酸、芦丁(峰3)、2-甲氧基-1,4-萘醌(峰7)、槲皮素(峰8)、山柰酚(峰14)5个峰。聚类分析与主成分分析结果基本一致,将10批药材聚为4类,PLS-DA表明,峰2、峰4、峰6、峰7、峰14是造成不同来源凤仙透骨草药材组间差异的标志性化合物;不同来源凤仙透骨草药材抗炎活性程度不同,变量重要性投影值表明峰2、峰4、峰6、峰7、峰8、峰14与抗炎活性具有较大的关联度。结论:建立了凤仙透骨草药材的指纹图谱,并确定了与抗炎活性密切相关的成分,可为凤仙透骨草药材质量评价提供参考依据。Objective:To establish the fingerprint of Fengxiantougucao(Impatiens balsamina L.)with high performance liquid chromatography(HPLC),and to explore the spectral-effect relationship with anti-inflammatory activity.Methods:The fingerprint of Fengxiantougucao was established with HPLC.The common peaks were evaluated by cluster analysis and principal component analysis.The carrageenan-induced acute inflammation models in mice were established to investigate the anti-inflammatory activity of Fengxiantougucao from different sources.The spectral-effect relationship was explored by using grey correlation analysis and partial least square discriminant analysis(PLS-DA).Results:The fingerprint of Fengxiantougucao was established,total 15 common peaks determined,and the similarity all above 0.90,and it also identified five peaks:ferulic acid,rutin(peak3),lawsone methyl ether(peak7),quercetin(pesk8),kaempferol(peak14).The results of cluster analysis and principal component analysis were basically the same.The 10 batches of medicinal materials were grouped into 4 categories.The results of PLS-DA showed that peaks 2,4,6,7,and 14 were responsible for the difference between the two groups.Different marker compounds had different anti-inflammatory activities from different sources.The variable importance in projection value indicated that peaks 2,4,6,7,8,and 14 had a greater degree of correlation with anti-inflammatory activity.Conclusions:The fingerprint of Fengxiantougucao has been established,and identified components closely related to anti-inflammatory activity,which can provide a reference for the quality evaluation of Fengxiantougucao.
