摘  要:目的研究胎球蛋白–A(fetoglobulin–a,Fetuin–A)调控糖尿病肾病足细胞的损伤机制。方法选取健康SD成年大鼠5只,获取大鼠肾小球基膜(glomerular basement membrane,GBM)足细胞,分为上调Fetuin–A组、下调Fetuin–A组。采用反转录–聚合酶链反应(real time polymerase chain reaction,RT–PCR)技术检测Fetuin–A、足细胞表达,观察细胞增殖、凋亡,免疫组化法检测肾组织肿瘤坏死因子–α(tumor necrosis factorα,TNF–α)、混合系列蛋白激酶结构域蛋白(mixed–lineage kinase domain–like protein,MLKL)、受体相互作用蛋白激酶1(receptor–interacting protein kinase 1,RIPK1)水平,蛋白质印迹(Western blot,WB)法检测Wilms瘤基因1(wilms,tumor gene 1,WT1)、特异蛋白1(specific protein 1,Sp1)表达水平。结果与对照组比较,下调Fetuin–A组Fetuin–A、TNF–α、MLKL、RIPK1表达水平、细胞吸光度值、细胞凋亡率均较低,WT1、Sp1表达水平较高;上调Fetuin–A组Fetuin–A、TNF–α、MLKL、RIPK1表达水平、细胞吸光密度值、细胞凋亡率均较高,WT1、Sp1表达水平较低(P<0.05);与下调Fetuin–A组比较,上调Fetuin–A组Fetuin–A、TNF–α、MLKL、RIPK1表达水平、细胞吸光度值、细胞凋亡率较高,WT1、Sp1表达水平较低(P<0.05)。结论Fetuin–A靶向作用于Sp1抑制足细胞生长失常。Objective To study the mechanism of fetoglobulin–A(Fetuin–A)regulating podocyte injury in diabetic nephropathy.Methods Five healthy SD adult rats were selected,and the glomerular basement membrane(GBM)podocytes of rats were obtained and divided into an up–regulated Fetuin–A group and a down–regulated Fetuin–A group.Polymerase chain reaction(real time polymerase chain reaction,RT–PCR)technique was used to detect Fetuin–A and podocyte expression,and cell proliferation and apoptosis were observed.Immunohistochemical method was used to detect tumor necrosis factor–α(TNF–α),mixed–lineage kinase domain–like protein(MLKL),receptor–interacting protein kinase 1(RIPK1)levels.Western blotting(WB)to detect the expression levels of Wilms tumor gene 1(WT1)and specific protein 1(Sp1).Results Compared with the control group,the Fetuin–A,TNF–α,MLKL,RIPK1 expression levels,cell absorbance density values,and cell apoptosis rate were lower in the Fetuin–A group down–regulated,while the expression levels of WT1 and Sp1 were higher in the Fetuin–A group.The expression levels of Fetuin–A,TNF–α,MLKL,RIPK1,cell absorbance density and apoptosis rate in the group were higher,while the expression levels of WT1 and Sp1 were lower(P<0.05).The expression levels of Fetuin–A,TNF–α,MLKL and RIPK1,cell absorbance density and apoptosis rate were higher in the up–regulated Fetuin–A group,while the expression levels of WT1 and Sp1 were lower(P<0.05).Conclusion Fetuin–a targets Sp1 to inhibit podocyte growth and dysfunctional.
