摘 要:目的观察褪黑素对大鼠挤压综合征模型骨骼肌缺血再灌注损伤的影响。方法将健康老年SD雄性大鼠用随机数字表法分4组,分为空白组(blank group,B组)、对照组(control group,C组)、氯化钠组(NaCl group,NS组)和褪黑素组(melatonin,MT组),予以5%戊巴比妥钠溶液,以50mg/kg剂量腹腔麻醉大鼠,利用卡式止血带挤压大鼠后肢根部,止血带拉力等同于3kg重物的压力,5h后撤除止血夹恢复挤压肢体血流灌注,其间大鼠能自由获取食物与水。空白组(B组)除腹腔给予5%戊巴比妥钠麻醉外无任何处理、对照组(C组)挤压肢体前无药物干预、氯化钠组(NS组)挤压肢体前30min予10ml/kg生理盐水和褪黑素组(MT组)挤压肢体前30min予10ml/kg褪黑素溶液,给药方式均通过大鼠尾静脉5min内推注。测定术后1~5d大鼠的游泳平均速度;于术前1d、术后1~5d进行Morris水迷宫实验测得逃避潜伏期、准确跨越平台次以及目标象限停留时间百分比,并于术后第6天测试其记忆能力;于术前1d、术后1、3、5d采集各组大鼠尾静脉血1ml,酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)检测血清中肿瘤坏死因子(tumor necrosis factor-alpha,TNF-α)、白细胞介素-10(interleukin-10,IL-10)水平。结果4组大鼠术后的游泳速度比较,差异无统计学意义(P>0.05);与B组比较,其他3组潜伏期显著延长(P<0.05);与对照组和氯化钠组比较,褪黑素组的潜伏期显著缩短(P<0.05),穿越平台次数显著增加(P<0.05),目标象限停留时间百分比显著增加(P<0.01),术后1、3、5d TNF-α、IL-10水平明显降低(P<0.05)。结论褪黑素对挤压综合征骨骼肌缺血再灌注损伤具有保护作用,并能提高挤压综合征模型大鼠的生存率。Objective To observe the effect of melatonin on skeletal muscle ischemia and reperfusion injury in rat squeeze syndrome models.Methods Healthy elderly male rats with SD were divided into four groups by random number table method,divided into blank group(group B),control group(group C),sodium chloride group(NS group)and melatonin group(MT group),used 5%sodium pentobarbital solution,intraperitoneal anesthesia rats at a dose of 50mg/kg,used cassette tourniquet to squeeze the root of the hind limbs of rats,the tourniquet pull equivalent to the pressure of 3kg weight,and after 5h,the hemostatic clip was removed to restore the perfusion of squeezed limb blood flow,during which the rats were free to obtain food and water.The blank group(group B)had no treatment except for intraperitoneal administration of 5%sodium pentobarbital anesthesia,no drug intervention before squeezing the limbs in the control group(group C),10ml/kg of normal saline for 30minutes before squeezing the limbs in the sodium chloride group(group C),and 10ml/kg melatonin solution for 30minutes before squeezing the limbs in the melatonin group(MT group),all of which were bolused through the rat tail vein within 5min.Determination of the average swimming speed of rats after surgery 1 to 5 days;Morris water maze experiments were performed 1 to 5 days before surgery to measure the evasion latency,accurate crossing of the platform and the percentage of dwell time in the target quadrant,and their memory ability was tested on the 6th day after surgery;1ml of tail vein blood in each group of rats was collected at 1 day before surgery,1 day after surgery,3 days and 5 days,and the levels of TNF-αand IL-10 were detected by enzyme linked immunosorbent assay,ELISA method.Results There was no significant difference in swimming speed after surgery among the four groups of rats(P>0.05);Compared with the other three groups,the incubation period was significantly longer(P<0.05),the incubation period was significantly shortened(P<0.05),the number of platform crossings w
