基于网络药理学研究桑白皮治疗类风湿性关节炎的作用机制

摘  要:应用网络药理学方法,研究桑白皮治疗类风湿性关节炎(RA)的作用机制.从TCMSP数据库中,查找出桑白皮活性成分及对应的作用靶点;结合GeneCards和DisGeNET数据库检索RA的相关靶点,并建立桑白皮治疗RA的活性成分及靶点网络;通过STRING数据库构建共有蛋白相互网络;最后利用DAVID6.8在线数据库进行GO功能分析和KEGG通路富集分析,并构建网络.筛选得到桑白皮有效成分共31个,如quercetin、kaempferol、Glabrone、Moracin D、Iristectorigenin(9CI)等,可调控的靶点有190个;RA靶点647个,桑白皮与RA的共同靶点有38个,主要有MAPK14、TNF、IKBKB、IL6等靶点;对关键靶点进行富集分析发现涉及桑白皮治疗RA的信号通路有肿瘤坏死因子信号通路、癌症通路等.桑白皮中的有效成分可能通过多个靶点作用于多条信号通路,从而发挥抗炎的作用.The mechanism of Morus alba L.in the treatment of rheumatoid arthritis was studied by network pharmacology.From TCMSP database,the active components and corresponding action targets of Morus alba L.were found;Relevant targets of rheumatoid arthritis were retrieved using GeneCards and DisGeNET databases,and an“active ingredient-target”regulatory network was constructed;Construction of protein protein interaction network through the STRING database;Finally,the DAVID6.8 online database was used for GO functional analysis and KEGG pathway enrichment analysis,and a network was constructed.A total of 31 active components of Morus alba L.were screened,such as quercetin,kaempferol,glabrone,moracin D,iristectorigenin(9CI),and 190 regulatable targets;There are 647 RA targets and 38 common targets of Morus alba L.and RA,mainly MAPK14,TNF,IKBKB,IL6 and other targets.The enrichment analysis of key targets found that the signal pathways involved in the treatment of RA by Morus alba L.include TNF signal pathway,cancer pathway and so on.The active components of Morus alba L.may act on multiple signal pathways through multiple targets,so as to play an anti-inflammatory role.
