升清降浊胶囊含药血清调控自噬减轻TGF-β1干预下大鼠系膜细胞纤维化的作用机制研究

摘　　要：目的:从自噬与肾纤维化角度初步探讨升清降浊胶囊含药血清对TGF-β干预下大鼠系膜细胞的影响。方法:以10 ng/ml的TGF-β为造模条件,制备不同浓度的含药血清对体外培养的大鼠肾小球系膜细胞(HBZY-1)进行干预,实验分为6组:对照组(CG),模型组(MG),升清降浊胶囊含药血清低剂量组(LSG)、中剂量组(MSG)、高剂量组(HSG),肾衰宁组(SNG)。CCK8法检测各组细胞活性的改变,MDC染色观察其自噬情况的变化,Western Blot检测α-SMA、CollagenⅣ、Beclin-1、LC3Ⅱ/Ⅰ蛋白表达的改变。结果:与CG组相比,MG组OD值增高(P<0.01),MDC阳性细胞百分比减低(P<0.01),α-SMA及CollagenⅣ表达增加(P<0.01),Beclin-1、LC3Ⅱ/Ⅰ表达减弱(P<0.01);与MG组相比,含药血清组OD值下降(P<0.01),MDC阳性细胞百分比增加(P<0.01),HSG组及SNG组CollagenⅣ(P<0.05)及α-SMA(P<0.01)表达减少,Beclin-1(P<0.01)、LC3Ⅱ/Ⅰ(P<0.05)表达增加。结论:升清降浊胶囊含药血清通过恢复系膜细胞自噬活性减轻TGF-β干预下大鼠系膜细胞纤维化的形成。Objective:To observe the effect of Shengqing Jiangzhuo Capsule Drug-containing serum on TGF-βinduced HBZY-1 rat mesangial cells from perspective of autophagy and renal fibrosis.Methods:HBZY-1 rat mesangial cells were induced by Transforming Growth Factor-beta 1(TGF-β)(10 ng/ml) and treated with different concentrations of Drug-containing serum.Cells were divided into:Control Group(CG),Model Group(MG),Low dose of Shengqing Jiangzhuo Capsule Drug-containing serum Group(LSG),Medium dose of Shengqing Jiangzhuo Capsule Drug-containing serum Group(MSG),High dose of Shengqing Jiangzhuo Capsule Drug-containing serum Group(HSG),Shengshuai Ning Capsule Drug-containing serum Group(SNG).CCK8 assay was performed to examine cell viability.MDC staining was used to observe the changes of cell autophagy.Western Blot method was used to observe the expression of α-SMA、Collagen Ⅳ、Beclin-1 and LC3Ⅱ/Ⅰ.Results:Compared with CG,OD value was increased in MG(P<0.01) and percentage of MDC positive cells was decreased,α-SMA and Collagen Ⅳ increased(P<0.01),Beclin-1 and LC3Ⅱ/Ⅰ(P<0.01) decreased.Compared with MG,OD value was decreased in Drug-containing serum groups(P<0.01) and percentage of MDC positive cells was increased,HSG and SNG Collagen Ⅳ(P<0.05) and α-SMA(P<0.01) decreased,Beclin-1(P<0.01) and LC3Ⅱ/Ⅰ(P<0.05) increased.Conclusion:Shengqing Jiangzhuo Capsules Drug-containing serum can reduce the TGF-βinduced fibrosis of HBZY-1 by promoting autophagy.
