摘 要:据报道Ⅰ群禽腺病毒(FAdV-I)在禽活疫苗中的污染是其可能的传播途径之一,加强对活疫苗中FAdV-I检测至关重要。为探索建立FAdV-I检测标准,本研究参照《中华人民共和国兽药典》中外源病毒检测相关疫苗处理措施,对不同方式处理后FAdV-I检测的影响进行了摸索。结果显示:当离心力不大于8000×g时,4℃离心10 min不会引起FAdV-I病毒含量下降,10000×g和12000×g分别离心10 min,病毒含量分别下降50%和68%,8000×g离心后再经0.45、0.22μm滤膜各滤过一次,病毒含量下降68%;8000×g离心后0.45、0.22、0.1μm滤膜各滤过1次后,病毒含量下降90%,12000×g离心后0.45、0.22、0.1μm滤膜各滤过1次后,病毒含量下降95%;SPF鸡血清及检验中常用的抗血清中和疫苗过程中在4℃或37℃作用1 h均不会引起FAdV-I病毒含量降低。上述指标为疫苗中FAdV-I污染的检测方法建立提供了参考数据。It has been reported that the contamination of subgroupⅠavian adenovirus(FAdV-I)in live avian vaccine is one of the possible transmissions.Therefore,it is important to strengthen the surveillance of FAdV-I in live vaccines.In order to develop reliable FAdV-I detection procedure,different processing methods were tried to detect FAdV-I contamination as referenced in the Veterinary Drug Code of the People's Republic of China.The results showed that the content of FAdV-I virus did not decrease after centrifugation at no more than 8000×g at 4℃for 10 minutes.However,the FAdV-I virus content decreased by 50%and 68%after centrifugation at 10000×g and 12000×g for 10 minutes,respectively.Moreover,centrifugation at 8000×g followed by fi ltration at 0.45 and 0.22μm decreased the virus content by 68%while fi ltration at 0.45,0.22 and 0.1μm decreased the virus content by 90%.When centrifugation increased to 12000×g followed by fi ltration at 0.45,0.22 and 0.1μm decreased the virus content by 95%.Treatment of vaccines with SPF chicken serum and commonly used antiserum at 4℃or 37℃for 1 hour did not cause the decrease of FAdV-I virus content.These data provided helpful information on the detection of FAdV-I contamination in live avian vaccines.
