摘 要:目的:通过抗惊厥作用和化学成分分析探讨僵蚕古代经典“粉末入药”的科学性与合理性。方法:建立戊四氮急性惊厥小鼠模型,以惊厥潜伏期、惊厥发生率、死亡潜伏期和存活率为指标评价僵蚕水煎液和粉末入药的抗惊厥作用差异;采用仿生提取法模拟粉末在胃肠环境中的溶出过程,通过干浸膏得率、蛋白类成分、白僵菌素含量和水溶性小分子成分测定,分析僵蚕古代经典“粉末入药”与现代“水煎入药”两种用药形式化学成分的差异,探讨僵蚕古代经典“粉末入药”的合理性。结果:僵蚕粉末入药对小鼠的抗惊厥作用显著强于水煎液;相比水煎液,僵蚕仿生液的干浸膏得率、蛋白含量明显增加;通过SDS-PAGE分析发现,僵蚕仿生液中蛋白类成分条带信息比水煎液明显丰富;HPLC分析结果表明,仿生液中水溶性成分组成更丰富,多数色谱峰峰面积明显增加,通过HPLC-Q-TOF-MS分析确定了其中部分成分的结构。结论:该研究证实了僵蚕古代经典“粉末入药”的科学合理性,可为僵蚕临床合理用药形式的选择提供科学依据。OBJECTIVE To explore the rationality of Bombyx Batryticatus(BB)used powder in clinic as recorded in ancient classic books based on the anticonvulsant effects and chemical composition analysis.METHODS By establishing the animal model of pentylenetetrazol-induced convulsion, the anticonvulsant effects of decoction and powder were evaluated by taking the seizure latency, seizure rate, death latency and survival rate as indexes.Bionic extraction method was used to simulate the dissolution process of powder in gastrointestinal environment, and taking the yield of dry extract, the change of protein components, the content of beauverine and water-soluble small molecular components as indicators, the differences of chemical components between the ancient classic medicinal form(powder)and the modern medicinal form(decoction)of BB for clinical application were analyzed.RESULTS The anticonvulsant effect of BB powder on mice was significantly stronger than that of decoction.compared with the decoction, the dry extract yield and protein content in biomimetic solution of BB were significantly increased.Through SDS-PAGE analysis, it was found that the information of protein components in the biomimetic solution of BB was obviously richer than that of the decoction.Through HPLC analysis, it was found that the water-soluble ingredients of the biomimetic solution were richer than that of the decoction, and the area of most chromatographic peaks was significantly increased.The structure of some ingredients was determined by HPLC-Q-TOF-MS.CONCLUSION The rationality of BB used as powder in clinic was confirmed in this study, which can provide a scientific basis for the rational use of BB in clinical practice.
