摘  要:目的:探索鹰嘴豆素A对IL-1β诱导的大鼠生长板软骨细胞凋亡的影响,同时对鹰嘴豆素A延迟大鼠骨骺闭合促进长骨生长的作用进行验证。方法:对4周龄SD大鼠生长板软骨细胞进行分离、培养及鉴定。正常培养的生长板软骨细胞为正常对照组,20 ng·mL^(-1)IL-1β诱导生长板软骨细胞凋亡为模型组,用药组在加入20 ng·mL^(-1)IL-1β的同时加入含有10μmol·L^(-1)鹰嘴豆素A的培养基进行培养,阳性对照组在加入20 ng·mL^(-1)IL-1β的同时加入5μmol·L^(-1)来曲唑进行培养。分别用MTT比色法和流式细胞仪检测鹰嘴豆素A抑制生长板软骨细胞凋亡的作用;同时以鹰嘴豆素A 100 mg·kg^(-1)给予大鼠灌胃,观察其对大鼠胫骨平台骨骺闭合及胫骨生长的影响。结果:MTT结果显示,与正常对照组相比,模型组软骨细胞增殖能力显著降低(P<0.01);与模型组相比,来曲唑和鹰嘴豆素A均能拮抗IL-1β对软骨细胞增殖的抑制作用,差异具有统计学意义(P<0.01),且鹰嘴豆素A的干预效果优于来曲唑;流式分析结果表明,鹰嘴豆素A能够降低IL-1β诱导的大鼠生长板软骨细胞的凋亡率,且干预效果优于阳性对照来曲唑;并且100 mg·kg^(-1)给予大鼠灌胃可以延迟大鼠胫骨平台骨骺闭合且促进胫骨生长。结论:鹰嘴豆素A对大鼠生长板软骨细胞凋亡抑制效果明显,可以延迟大鼠胫骨平台的骨骺闭合,为骨骼生长争取更长时间,从而促进发育期骨骼生长。OBJECTIVE To explore the effect of biochanin A on the apoptosis of growth plate chondrocytes induced by IL-1β,and verify the effect of biochanin A in delaying the closure of the rat epiphysis and promoting the growth of long bones.METHODS The growth plate chondrocytes of 4-week-old SD rats were isolated and cultured.Growth plate chondrocytes in the normal control group were normally cultured,and growth plate chondrocytes in the model group were induced by 20 ng·mL^(-1)of IL-1β,20 ng·mL^(-1)IL-1βand medium containing 10μmol·L^(-1)biochanin A were added to the experimental group of cells,20 ng·mL^(-1)of IL-1βand 5μmol·L^(-1)of letrozole were added to the positive control group.The apoptosis of chondrocytes in each group was detected by MTT colorimetric assay and flow cytometry;biochanin A was used for gastric lavage in rats,and its effect on the closure of the epiphyseal tibial plateau and the growth of the tibia was observed.RESULTS The proliferation ability of chondrocytes in the model group was significantly reduced compared with the normal control group(P<0.01);Compared with the model group,letrozole and biochanin A could antagonize the inhibitory effect of IL-1βon chondrocyte colonization,and the difference was statistically significant(P<0.01).The intervention effect of biochanin A was better than that of letrozole.The results of flow cytometry analysis showed that biochanin A could reduce the apoptosis rate of IL-1β-induced rat growth plate chondrocytes,and the intervention effect was better than that of positive control group;Administration of biochanin A at a concentration of 100 mg·kg^(-1)to rats could delay the closure of the epiphyseal tibial plateau in rats and promote tibia growth.CONCLUSION Biochanin A had a significant inhibitory effect on apoptosis of rat growth plate chondrocytes and could delay epiphyseal closure of the tibia plateau in rats,thereby providing longer periods of bone growth and promoting the growth of bones during development.
