摘 要:采用ITS、ITS2、psbA-trnH、rbcL和matK等序列对我国口岸20种菊科杂草籽进行扩增测序,比较各序列的扩增和鉴定成功率,采用最佳鉴定序列扩增6种检疫性菊科杂草籽序列,计算6种菊科杂草籽种间K2P距离并构建与其近似种的系统进化树。结果表明,ITS2扩增和鉴定成功率最高,说明利用DNA条形码能够准确地鉴别菊科杂草,ITS2序列是鉴别菊科杂草较理想的条形码序列。同时通过智能3D数码显微镜,采集5种菊科杂草籽形态学特征,形成形态学图谱。该研究结果为海关一线关员鉴定检疫性菊科杂草提供了参照依据,也为外来有害生物鉴定提供了科学依据与新的思路。The sequences of ITS,ITS2,psbA-trnH,rbcL and matK were used to amplify and sequence 20 species of Compositae weed seeds at Chinese ports,and the amplification and sequencing efficiency of each sequence were compared.The K2P distance and NJ tree of the 6 quarantine compositae weeds were analyzed by using the best identification sequence.According to the results,the amplification and sequencing efficiencies of ITS2 were best.ITS2 could be used as a potential DNA barcode combination to correctly identify the Compositae weeds.At the same time,the morphological characteristics of 5 Compositae seeds were collected to form morphological map by VHX-6000.The research provides a reference for the front-line Customs officers to identify quarantine Compositae weeds,and also provides a scientific basis and a new idea for the identification of exotic pests.
