摘 要:目的:探讨减味寿胎丸对流产大鼠母胎界面中叉头框转录因子3(FOXO3)、孕激素受体(PR)表达的影响。方法:35只SPF级雌性SD大鼠,妊娠第0天随机分为5组:正常组,模型组,地屈孕酮组,减味寿胎丸低、高剂量(JWL、JWH)组,每组7只。碘[125I]人绒毛膜促性腺激素放射免疫分析法检测血清孕激素(P)、雌激素(E2)、大鼠游离β绒毛膜促性腺激素(f-βCG)水平,体视镜观察孕鼠子宫、胚胎,HE染色观察子宫胚胎结合部位,RT-qPCR检测子宫胚胎结合部位FOXO3、PR mRNA表达,Western blot检测子宫胚胎结合部位FOXO3、PR蛋白表达。结果:模型组孕鼠出现肾虚宏观表现,流产率显著高于正常组(P<0.01),与模型组比较,JWL、JWH组孕鼠流产率均显著降低(P<0.05)。与正常组比较,模型组孕鼠血清P、E2、f-βCG水平显著降低(P<0.05);模型组孕鼠子宫胚胎结合部组织中FOXO3、PR mRNA及蛋白表达显著降低(P<0.05,P<0.01);与模型组比较,JWL组、JWH组各指标均显著升高(P<0.05,P<0.01)。结论:减味寿胎丸可降低流产大鼠模型流产率,改善胚胎供血,其作用机制可能是通过上调母胎界面潜在靶基因FOXO3从而促进下游PR的表达,维持孕期黄体功能,达到补肾安胎的作用。Objective: To investigate the effects of Jianwei Shoutai Pills of FOXO3 and progesterone receptor(PR)expression in the maternal-fetal interface of abortion model rats. Methods: Thirty-five SPF female SD rats were randomly divided into 5 groups on the 0th day of pregnancy: control group, model group, dydrogesterone group, Jianwei Shoutai Pills low-dose(JWL) group, Jianwei Shoutai Pills high-dose(JWH) group, 7 rats in each group. 125I radioimmunoassay was used to detect serum progesterone(P), estrogen(E2), rat free beta chorionic gonadotropin(f-βCG) levels, stereoscope was used to observe uterus and embryos of pregnant rats, HE staining was used to observe uterine-embryo binding location, RT-qPCR was used to detect the expression of FOXO3 and PR mRNA in the uterine embryo binding site, and Western blot was used to detect the expression of FOXO3 and PR protein in the uterine embryo binding site. Results: The model group showed macroscopic manifestations of kidney deficiency, and the abortion rate in the model group was higher than that in the normal group(P<0.01). Compared with the model group, the abortion rates in the JWL and JWH groups were significantly lower(P<0.05). Compared with the normal group,the serum levels of P, E2 and f-βCG in the pregnant mice of the model group were significantly decreased(P<0.05);the mRNA and protein expressions of FOXO3 and PR in the junction of the uterus and embryos of the pregnant mice in the model group were significantly decreased(P<0.05, P<0.01);Compared with the model group, all indexes in JWL group and JWH group were significantly increased(P<0.05, P<0.01). Conclusion: Jianwei Shoutai Pills can reduce the abortion rate and improve the blood supply of the embryo in the abortion rat. Its mechanism of action may be through up-regulating the potential target gene FOXO3 at the maternal-fetal interface to promote the expression of downstream PR, maintain the function of the corpus luteum during pregnancy, and achieve the effect of invigorating the kidney and relieving the fetus
