摘  要:目的 探讨壮药白花九里明提取物绿原酸含量测定方法及其对早孕子宫出血模型大鼠心肌细胞的影响。方法 采用高效液相色谱法(HPLC)测定白花九里明提取物绿原酸的含量;建立早孕子宫出血大鼠模型,分组灌药7 d后,取心肌组织固定,包埋,苏木精-伊红(HE)染色,切片。结果 白花九里明提取物中绿原酸的检测波长为330 nm、流动相为乙腈-0.1%磷酸(8∶92),绿原酸在0.24~1.20μg质量范围内与其对应的峰面积呈良好的线性关系,回归方程为y=2 000 000x+6 159.6,R^(2)=0.999 3,平均回收率为97.89%,相对标准偏差(RSD)=1.36%,白花九里明提取物中绿原酸含量为(4.485 0±0.062 5)mg/g;早孕子宫出血大鼠模型心肌细胞无明显变化。结论 白花九里明提取物中含绿原酸,对早孕子宫出血大鼠模型心肌细胞无影响,提示白花九里明提取物对动物模型用药心脏安全。Objective To investigate the Blumea megacephala(Randeria) Chang et Tseng of chlorogenic acid on myocardium of rats with early pregnancy uterine bleeding.Methods The content of chlorogenic acid in the extract of Blumea megacephala(Randeria) Chang et Tseng was determined by high performance liquid chromatography(HPLC).The rat model of uterine bleeding in early pregnancy was established.After 7 days of administration of chlorogenil acid, the myocardial tissue was fixed, embedded, stained with hematoxylin-eosin(HE) and observed.Results The detection wavelength of chlorogenic acid in Blumea megacephala(Randeria) Chang et Tseng extract was 330 nm, and the mobile phase was acetonitrile-0.1% phosphoric acid(8∶92). There was a good linear relationship between chlorogenic acid and its corresponding peak area in the mass range of 0.24-1.2 μg.The regression equation was y=2 000 000x+6 159.6,R^(2)=0.999 3,the average recovery was 97.89%,relative standard deviation(RSD) was 1.36%,and the content of chlorogenic acid in Blumea megacephala(Randeria) Chang et Tseng extract was(4.485 0±0.062 5) mg/g.There was no significant change in cardiomyocytes in the rat model of uterine bleeding in early pregnancy.Conclusion The extract of Blumea megacephala(Randeria) Chang et Tseng contains chlorogenic acid, which has no effect on cardiomyocytes of early pregnant rats with utrine bleeding, suggesting that the extract is safe for the heart of animal models.
