摘  要:目的:利用超高效液相色谱-四极杆-静电场轨道阱质谱法(UPLC-Q-Exactive-Orbitrap-MS)对苗族药刺梨叶中的化学成分进行准确、快速的定性鉴别。方法:采用Waters ACQUITY UPLC HSS T3 C_(18)色谱柱(150 mm×2.1 mm,1.7µm),以0.1%甲酸水溶液-乙腈为流动相进行梯度洗脱,流速为0.3 mL·min^(-1),柱温40℃,进样量2µL。电喷雾离子源(ESI)在正、负离子2种模式下采集数据。扫描方式为全扫描/数据依赖二级扫描(Full MS/dd-MS2)。结果:在刺梨叶的75%甲醇提取液中共鉴定出57个化学成分,包括黄酮类21个、氨基酸类12个、有机酸类10个、多酚类5个、三萜类3个、植物甾醇类2个、蒽醌类1个、四环二萜类1个、香豆素类1个及其他类化合物1个。结论:采取UPLC-Q-Exactive-Orbitrap-MS快速有效地鉴定出苗族药刺梨叶中化学成分57个,为进一步明确刺梨叶的药效物质基础及完善其质量标准提供参考。Objective:To qualitatively analyze the chemical components of Rosae Roxburghii Folium based on UPLC-Q-Exactive-Orbitrap-MS.Methods:UPLC conditions are as follows:Waters ACQUITY UPLC HSS T3 C_(18) column(150 mm×2.1 mm,1.7µm),gradient elution with 0.1%formic acid aqueous solution-acetonitrile as the mobile phase,flow rate of 0.3 mL·min^(-1),column temperature of 40℃,and injection volume of 2µL.MS conditions are as below:ESI+/−and full mass scan and data-dependent MS/MS(Full MS/dd-MS2).Results:A total of 57 chemical compounds were identified in 75%methanol extract of Rosae Roxburghii Folium:21 flavonoids,12 amino acids,10 organic acids,5 polyphenols,3 triterpenoids,2 phytosterols,1 anthraquinone,1 tetracyclic diterpenoid,1 coumarin,and 1 miscellaneous.Conclusion:UPLC-Q-Exactive-Orbitrap-MS was used to quickly and effectively identify 57 components in Rosae Roxburghii Folium.The findings can serve as a reference for further clarifying the therapeutic material basis of Rosae Roxburghii Folium and improving its quality standard.
