摘  要:目的探讨黄芩苷(baicalin)对脂多糖(lipopolysaccharide,LPS)诱导的人单核细胞THP-1的炎症反应的作用。方法利用LPS建立人单核细胞THP-1的炎症模型,实时荧光定量聚合酶链式反应(real-time quantitatFive polymerase chain reaction,RT-qPCR)方法检测白细胞介素-1β(interleukin-1β,IL-1β)、IL-6、IL-8、粒细胞-巨噬细胞集落刺激因子(granulocyte-macrophage colony-stimulating factor,GM-CSF)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的相对表达,确定最优的LPS刺激浓度。乳酸脱氢酶检测试剂盒检测黄芩苷的细胞毒性,优选其最佳使用浓度范围。对LPS诱导的人单核细胞THP-1炎症模型予以不同浓度的黄芩苷预处理,利用RT-qPCR法检测黄芩苷对LPS诱导的人单核细胞THP-1的IL-1β、IL-6、IL-8、GM-CSF和TNF-α的表达升高的影响。结果与对照组相比,1μg/mL的LPS处理人单核细胞THP-124 h后均可显著引起IL-1β、IL-6、IL-8、GM-CSF和TNF-α的表达升高(P<0.05),黄芩苷在浓度为10、30、100μmol/L时处理人单核细胞THP-124 h和48 h,无明显细胞毒性。30μmol/L和100μmol/L的黄芩苷可降低LPS诱导的人单核细胞THP-1的IL-1β、IL-6、IL-8、GM-CSF和TNF-α的表达升高。结论黄芩苷可显著抑制LPS诱导的人单核细胞THP-1的炎症反应。Objective To observe the effect of baicalin on the lipopolysaccharide(LPS)-stimulated inflammatory response in human monocytes THP-1.Methods LPS was used to induce the inflammatory response of human monocyte THP-1,and the relative expression levels of interleukin-1β(IL-1β),IL-6,IL-8,granulocyte-macrophage colony-stimulating factor(GM-CSF)and tumor necrosis factor-α(TNF-α)were detected by real-time quantitative polymerase chain reaction(RT-qPCR)method to optimize the stimulation concentration of LPS.The cytotoxic effects of baicalein were also detected using the lactate dehydrogenase assay kit to optimize the bacailin concentration.The LPS-induced inflammation model of human monocyte THP-1 was pretreated with baicalin at different concentrations,and the inhibition effect of baicalin on the increasing expression of IL-1β,IL-6,IL-8,GM-CSF and TNF-αinduced by LPS was examined by RT-qPCR.Results Compared to the control,the LPS treatment of human monocytes THP-1 at 1μg/mL for 24 h could significantly increase the expression of IL-1β,IL-6,IL-8,GM-CSF and TNF-α(P<0.05).Treatment with baicalin at 10,30 and 100μmol/L on human monocytes THP-1 for 24 h and 48 h showed no significant cell cytotoxicity.The baicalein at 30μmol/L and 100μmol/L could significantly inhibit the LPS-induced increasing expression of IL-1β,IL-6,IL-8,GM-CSF and TNF-αin human monocytes THP-1.Conclusion The baicalin could significantly inhibit LPS-induced inflammatory response in human monocytes THP-1.
