摘  要:苯甲醛苯甲酰腙类化合物具有一定的农药应用价值,但也可能对人体产生不良影响。借助荧光光谱技术和分子模拟技术,研究邻硝基苯甲醛2,4-二氯苯甲酰腙(NCH)与人血清白蛋白(HAS)之间的相互作用模式与结合方式,以期为新型农药的开发提供参考。荧光猝灭实验结果表明:NCH可使HSA发生荧光猝灭,且其猝灭机制为静态猝灭;相关热力学参数ΔS>0,ΔH<0,说明NCH与HSA之间的作用力以疏水作用力为主,但氢键和范德华作用力也不可忽视。位点竞争实验和分子模拟结果表明:NCH与HSA的结合部位为HSA的SiteⅠ部位。同步荧光光谱实验结果表明:NCH使HSA酪氨酸微环境发生变化,说明NCH改变了HSA的构象。Benzaldehyde benzoyl hydrazone compounds have certain value in pesticide applications,but they may cause adverse effects on the human body.In this paper,the fluorescence spectroscopy technology and molecular simulation technology were used to study the interaction patterns and bonding patterns between o-nitrobenzaldehyde 2,4-dichlorobenzoylhydrazone(NCH)and human serum albumin(HSA),in order to provide some reference for the development of new pesticides.Fluorescence quenching experiment results showed that NCH could quench the fluorescence of HSA.The related thermodynamic parametersΔS>0 andΔH<0 indicated that the force between NCH and HSA was mainly hydrophobic,but the hydrogen bond and van der Waals force could not be ignored.The results of site competition experiment and molecular simulation showed that the binding site of NCH and HSA was Site I of HSA.The experimental results of synchronous fluorescence spectroscopy showed that NCH changed the conformation of HSA.The results of synchronous fluorescence spectroscopy experiments showed that NCH changed the HSA tyrosine microenvironment,indicating that NCH changed the conformation of HSA.
