摘  要:目的 研究一种刺五加提取物对人神经母细胞瘤细胞SH-SY5Y氧糖剥夺再灌注(Oxygen and Glucose Deprivation/Reperfusion,OGD/R)后损伤的影响。方法 体外培养SH-SY5Y细胞,并分成空白对照组(Natural Control,NC)、氧糖剥夺再灌注模型组(OGD/R)、阳性对照组(80μM依达拉奉Edaravone+OGD/R)、刺五加提取物中剂量组(500μg·mL^(-1)+OGD/R)、刺五加提取物高剂量组(1000μg·mL^(-1)+OGD/R)。其中,NC组给予DMEM/F12培养基培养,模型组给予氧糖剥夺6 h、复氧复糖再培养12 h处理。各组采用MTT法检测细胞活力,流式细胞术检测凋亡水平,多功能酶标仪及荧光显微镜检测活性氧(ROS)、超氧化物歧化酶(SOD)、丙二醛含量(MDA)。结果 与模型组相比,刺五加提取物能显著降低OGD/R引起的SH-SY5Y细胞凋亡;降低活性氧及丙二醛水平,提高超氧化物歧化酶活力,提高胞内ATP水平。结论 刺五加提取物可通过抑制氧化应激、保护线粒体功能和抑制细胞凋亡来减轻SH-SY5Y细胞氧糖剥夺后再灌注损伤的作用。Objective To study the effects of Acanthopanax senticosus extract on the injury of SH-SY5Y cells after oxygen-glucose deprivation and reperfusion(OGD/R).Methods SH-SY5Y cells were cultured in vitro and divided into blank Control group(Natural Control, NC), oxygen and glucose deprivation reperfusion model group(OGD/R), positive control group(80 μM Edaravone+ OGD/R), middle dose group of Acanthopanax senticosus extract(500 μg·mL^(-1)+ OGD/R), high dose group of Acanthopanax senticosus extract(1000 μg·mL^(-1)+ OGD/R). The NC group was cultured with DMEM/F12 medium, and the model group was treated with oxygen and glucose deprivation for 6 hours and reoxygenation and reoxygenation for 12 hours. MTT assay was used to detect the cell viability;flow cytometry was used to detect the apoptosis level;multifunctional microplate analyzer and fluorescence microscope were used to detect the content of reactive oxygen species(ROS), superoxide dismutase(SOD) and malondialdehyde(MDA). Results Compared with model group(OGD), Acanthopanax senticosus extract significantly reduced the apoptosis of SH-SY5Y cells induced by OGD/R. It decreased the level of reactive oxygen species and malondialdehyde, increased the activity of superoxide dismutase, increased the level of intracellular ATP.Conclusion The extract of Acanthopanax senticosus can reduce the reperfusion injury of SH-SY5Y cells after oxygen and sugar deprivation by inhibiting oxidative stress, protecting mitochondrial function and inhibiting apoptosis.
