摘  要:目的探讨橙皮素是否通过调节氧化应激及细胞凋亡对糖尿病兔心肌缺血再灌注损伤(MIRI)发挥保护作用。方法雄性家兔24只,随机分为对照组、糖尿病心肌缺血再灌注损伤(D-MIRI)组、低剂量橙皮素+D-MIRI(L-HSP+D-MIRI)组、高剂量橙皮素+D-MIRI(H-HSP+D-MIRI)组,每组6只。应用超氧化物歧化酶(SOD)和丙二醛(MDA)试剂盒检测4组兔心肌组织中SOD和MDA水平。苏木素-伊红(HE)染色观察心肌组织病理变化。末端DNA转移酶dUPT缺口末端标记(TUNEL)法检测兔心肌组织细胞凋亡变化。结果L-HSP+D-MIRI组、H-HSP+D-MIRI组心肌组织损伤轻于D-MIRI组。H-HSP+D-MIRI组心肌组织损伤程度轻于L-HSP+D-MIRI组。L-HSP+D-MIRI组、H-HSP+D-MIRI组心肌组织SOD水平高于D-MIRI组(P<0.05)。H-HSP+D-MIRI组心肌组织SOD水平高于L-HSP+D-MIRI组(P<0.05)。L-HSP+D-MIRI组、H-HSP+D-MIRI组心肌组织MDA水平和细胞凋亡率低于D-MIRI组(P<0.05)。H-HSP+D-MIRI组心肌组织MDA水平和细胞凋亡率低于L-HSP+D-MIRI组(P<0.05)。结论橙皮素通过抑制心肌组织氧化应激和细胞凋亡对兔D-MIRI发挥保护作用。Objective To investigate the protective effect of hesperidin on myocardial ischemia-reperfusion injury(MIRI)in diabetic rabbits by regulating oxidative stress and apoptosis.Methods Twenty-four male rabbits were randomly divided into the control group,diabetic-D-MIRI group(D-MIRI),low-dose of hesperidin+D-MIRI group(L-HSP+D-MIRI),and high-dose of hesperidin+D-MIRI group(H-HSP+D-MIRI),6 cases in each group.The levels of SOD and MDA in myocardial tissue of the four groups were detected by using the SOD and MDA kits.The pathological changes of myocardial tissue were observed by HE staining.TUNEL assay was used to detect the cell apoptosis of rabbit myocardial tissue.Results The myocardial tissue damage in the L-HSP+D-MIRI group was lighter than that in the D-MIRI group.The degree of myocardial tissue injury in the H-HSP+D-MIRI group was lighter than that in the L-HSP+D-MIRI group.The level of SOD of myocardial tissue in the L-HSP+D-MIRI group was higher than that in the D-MIRI group(P<0.05).The level of SOD of myocardial tissue in the H-HSP+D-MIRI group was higher than that in the L-HSP+D-MIRI group(P<0.05).The level of MDA and apoptosis rate of myocardial cells in the L-HSP+D-MIRI and H-HSP-D-MIRI group was lower than those in the D-MIRI group(P<0.05).The MDA level of the myocardial tissue and apoptosis of myocardial cells rate in the H-HSP+D-MIRI group was lower than those in the L-HSP+D-MIRI group(P<0.05).Conclusion Hesperidin plays a protective role on D-MIRI of rabbit myocardium by inhibiting oxidative stress and apoptosis of myocardial cells.
