摘 要:本研究旨在开发一种准确快速鉴别红景天属植物的方法。选用14种红景天属植物进行ITS2和matK序列的测定,并对其进行序列特征和系统发育分析。ITS2和matK条形码序列的扩增成功率均为100%。ITS2序列测序成功率为94.7%,matK序列的测序成功率为100%。系统发育分析表明ITS2序列能够区分大花红景天、西藏红景天和长鞭红景天,但是不能把柴胡红景天和狭叶红景天区分开;matK序列能够区分大花红景天和柴胡红景天,但是不能把西藏红景天和长鞭红景天区分开,也不能把异鳞红景天、长毛圣地红景天与互生红景天区分开。基于ITS2和matK序列联合构建的系统发育树能够区分14种红景天属植物。以ITS2和matK序列组成联合序列的DNA条形码鉴定方法能够对14种红景天植物进行快速准确的鉴定。The aim of this study was to develop an accurate and rapid identification method for Rhodiola species.Forteen Rhodiola plants were selected for the determination of ITS2 and matK sequences,and their sequence characteristics and phylogenetic analysis were carried out.The amplification success rate of both ITS2 and matK barcode sequences was 100%.The success rate of ITS2 sequence sequencing was 94.7%,and the sequencing success rate of matK sequence was 100%.Phylogenetic analysis showed that the ITS2 sequence could distinguish Rhodiola crenulata,Rhodiola tibetica and Rhodiola fastigiata,but could not distinguish Rhodiola bupleuroides and Rhodiola kirilowii.The matK sequence could distinguish Rhodiola crenulata and Rhodiola bupleuroides,but could not distinguish between Rhodiola tibetica and Rhodiola fastigiata,nor could it distinguish Rhodiola smithii,Rhodiola sacra,and Rhodiola alterna.The phylogenetic tree based on the combined construction of ITS2 and matK sequences was able to distinguish fourteen Rhodiola plants.The DNA barcode identification method with the combined sequence of ITS2and matK sequences was able to identify fourteen Rhodiola plants quickly and accurately.
