毛果杨接种双色蜡蘑培养外生菌根的方法及其对毛果杨生长的影响

摘  要:以无菌组培毛果杨(Populus trichocarpa)幼苗为宿主植物,以无菌石英砂为基质,接种纯培养双色蜡蘑(Laccaria bicolor),发现毛果杨幼苗与外生菌根真菌双色蜡蘑在无菌环境中形成了明显的外生菌根结构,在毛果杨—双色蜡蘑共聚焦显微镜横切片中观察到典型的哈氏网.此外,相较于不接种双色蜡蘑、添加培养液的毛果杨幼苗,接种了双色蜡蘑的毛果杨幼苗的株高增长,鲜重显著增加.说明外生菌根真菌双色蜡蘑能促进毛果杨幼苗的生长.Laccaria bicolor was used to inoculate tissue culture plantlets of Populus trichocarpa grown in sterile quartz sand,and the developments of ectomycorrhiza and P.trichocarpa plantlet were further investigated.The results showed that obvious ectomycorrhizal structures were formed between P.trichocarpa plantlet and L.bicolor in the sterile environment,and simultaneously typical Hartig net structures were observed in the cross-section of ectomycorrhiza under laser scanning confocal microscopy.In addition,compared with the control,P.trichocarpa plantlets inoculated with L.bicolor had higher plant height and significantly higher fresh weight,indicating the promotion of ectomycorrhizal fungus L.bicolor on the growth of P.trichocarpa plantlet.
