摘  要:建立了HPLC内标法同时测定姜黄油树脂中双去甲氧基姜黄素、去甲氧基姜黄素、姜黄素、芳香姜黄酮、α-姜黄酮含量的方法。采用Thermo C_(18)色谱柱(250 mm×4.6 mm,5.0μm),流动相为含0.1%冰醋酸的乙腈(A)-0.1%冰醋酸水溶液(B),梯度洗脱,流速1.0 mL·min^(-1),柱温35℃,检测波长分别为420 nm(0~8 min)、238 nm(8~15 min)、262 nm(15~40 min)。双去甲氧基姜黄素、去甲氧基姜黄素、姜黄素、芳香姜黄酮、α-姜黄酮分别在1.388×10^(-2)~3.553μg(r=0.9999)、1.395×10^(-2)~3.570μg(r=1.0000)、1.382×10^(-2)~3.539μg(r=0.9998)、1.388×10^(-2)~3.554μg(r=0.9999)、1.719×10^(-2)~4.400μg(r=0.9997)的范围内与色谱峰峰面积呈良好的的线性关系;平均回收率(n=9)分别为98.0%、98.1%、101.1%、99.5%、98.7%,RSD分别为2.3%、2.8%、2.0%、0.7%、1.6%。经分析测定,在被检测的6批姜黄油树脂样品中双去甲氧基姜黄素、去甲氧基姜黄素、姜黄素、芳香姜黄酮、α-姜黄酮的含量测定结果分别为15.454~28.132、14.604~18.088、8.446~9.881、31.701~41.128、3.496~4.425μg·mL^(-1)。该方法准确、灵敏、操作简便,可用于姜黄油树脂中双去甲氧基姜黄素、去甲氧基姜黄素、姜黄素、芳香姜黄酮、α-姜黄酮的含量测定。An HPLC internal standard method was established for simultaneous determination of bisdemethoxylcurcumin,demethoxylcurcumin,curcumin,ar-Turmerone andα-Turmerone in Turmeric oleoresin.The analysis was performed on a Thermo C_(18) column(250 mm×4.6 mm,5.0μm),using acetonitrile containing 0.1%glacial acetic acid(A)-0.1%glacial acetic acid solution(B)as mobile phase under gradient elution at flow rate 1.0mL·min^(-1).The column temperature was 35℃and the detection wavelengths were 420 nm(0~8 min)、238 nm(8~15 min)、262 nm(15~40 min).There was a good linear relationship in the ranges of 1.388×10^(-2)~3.553μg(r=0.9999)for bisdemethoxylcurcumin,1.395×10^(-2)~3.570μg(r=1.0000)for demethoxylcurcumin,1.382×10^(-2)~3.539μg(r=0.9998)for curcumin,1.388×10^(-2)~3.554μg(r=0.9999)for ar-Turmerone and 1.719×10^(-2)~4.400μg(r=0.9997)forα-Turmerone.The average recoveries were 98.0%,98.1%,101.1%,99.5%,and 98.7%with RSDs of 2.3%,2.8%,2.0%,0.7%and 1.6%,respectively.The contents of above-mentioned compounds in the 6 batches of Turmeric oleoresin were determined as 15.454~28.132μg·mL^(-1) for bisdemethoxylcurcumin,14.604~18.088μg·mL^(-1) for demethoxylcurcumin,8.446~9.881μg·mL^(-1) for curcumin,31.701~41.128μg·mL^(-1) for ar-Turmerone and 3.496~4.425μg·mL^(-1) forα-Turmerone.The method is accurate,sensitive and simple,which is suitable for the determination of bisdemethoxylcurcumin,demethoxylcurcumin,curcumin,ar-Turmerone andα-Turmerone in Turmeric oleoresin.
