摘 要:针对条斑紫菜(Pyropia yezoensis)DnaJ(PyDnaJ)家族研究相对较少的现状,本研究利用生物信息学方法对PyDnaJ基因家族及其在胁迫条件下的表达模式进行探究,以便为进一步解析条斑紫菜抗逆机制奠定前期工作基础。本研究鉴定出27个DnaJ蛋白(PyDnaJ01~27),并发现:其编码氨基酸数量范围为191~980;大部分DnaJ定位于高尔基体和细胞核;成员的基因结构简单,内含子较少;根据所含结构域不同可分为A、B、C三类;27个成员散布于3条染色体上。其中:PY_1染色体含DnaJ基因最多(11个);PyDnaJ01和PyDnaJ10为片段重复基因;PY_2和PY_3染色体上各有8个DnaJ基因;DnaJ基因启动子区域含有不同数目的胁迫响应元件和植物激素响应元件。系统发生分析表明PyDnaJ蛋白家族可以分成10个亚群。qRT-PCR结果显示,PyDnaJ家族成员在孢子体世代下的表达水平显著高于配子体世代,PyDnaJ家族成员在失水及温度胁迫下具有不同表达模式。In view of the relatively few studies on Pyropia yezoensis DnaJ(PyDnaJ)family,this study explored the PyDnaJ gene family and its expression pattern under stress conditions by bioinformatics methods,in order to lay a foundation for further analysis of stress resistance mechanism of P.yezoensis.In this study,we identified 27 members(PyDnaJ01-27).Results showed that the number of amino acids of 27 DnaJ proteins was varied from 191 to 980,and most DnaJ proteins were located in golgi apparatus and nucleus.The gene structure of PyDnaJ was relatively simple,with fewer introns.27 PyDnaJ proteins could be classified into three types of A,B and C.27 gene family members were scattered on 3 chromosomes.PY_1 contained 11 DnaJ genes,among which PyDnaJ01 and PyDnaJ10 were segmental duplications.PY_2 and PY_3 each contained 8 DnaJ genes.The promoter regions of DnaJ genes contained different numbers of stress response elements and plant hormone response elements.Evolutionary analysis showed that PyDnaJ proteins could be distinguished into 10 sub-clusters.The results of qRT-PCR showed that expression level of PyDnaJ family members in the sporophyte generation was significantly higher than that in the gametophyte generation.Moreover,PyDnaJ family members showed different expression patterns under water loss and temperature stress.This study laid a foundation for further analysis of the stress resistance mechanism of P.yezoensis.
