摘  要:目的:通过高嘌呤饮食构建果蝇高尿酸血症模型,探讨尿酸(UA)对果蝇生长发育和寿命的影响及机制。方法:收集羽化后8 h内的果蝇1000只,随机分为对照组(0%腺嘌呤干预)、低浓度干预组(0.1%腺嘌呤干预)、中浓度干预组(0.2%腺嘌呤干预)和高浓度干预组(0.3%腺嘌呤干预)。观察各组果蝇的寿命、攀爬能力及产卵量情况,并应用ELISA测定果蝇体内UA及相关酶的变化,以及氧化应激相关指标的表达变化。此外,收集果蝇卵400只,与上述分组相同,观察各组幼虫的蛹化及羽化情况。结果:与对照组比较,随着培养基中腺嘌呤浓度的升高,各干预组果蝇体内UA、尿酸氧化酶(UOD)和黄嘌呤氧化酶(XOD)表达水平显著升高(P<0.01)。各干预组超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)表达水平及总抗氧化能力(T-AOC)显著升高(P<0.01),一氧化氮(NO)水平显著降低(P<0.01),而丙二醛(MDA)水平则呈现先升高后降低的趋势。与对照组比较,随着果蝇体内UA水平的不断升高,各干预组果蝇的平均寿命显著降低(P<0.01),中、高浓度干预组雌性果蝇的产卵量和攀爬能力显著降低(P<0.01),而各干预组雄性果蝇的攀爬能力均显著降低(P<0.01)。与对照组比较,低浓度干预组果蝇幼虫的蛹化率及羽化率显著降低(P<0.01)。结论:高浓度UA抑制果蝇的生长发育,降低果蝇的寿命、产卵量及攀爬能力,其机制可能与果蝇体内氧化-抗氧化系统失衡有关。AIM:The hyperuricemia model of Drosophila melanogaster was established by using high purine diet to investigate the effects of uric acid(UA)on growth and life span of Drosophila melanogaster and its mechanism.METHODS:A total of 1000 Drosophila were collected within 8 h after eclosion and randomly divided into four groups:control group(0%-adenine treated),low concentration intervention group(0.1%-adenine treated),medium concentration intervention group(0.2%-adenine treated)and high concentration intervention group(0.3%-adenine treated).The life span,climbing ability and oviposition quantity of each group were observed,and the changes of UA and related enzymes,as well as the expression of oxidative stress related indexes in Drosophila were determined by ELISA.In addition,400 Drosophila eggs were collected,and the pupation and emergence of larvae in each group were observed.RESULTS:Compared with the control group,UA,urate oxidase(UOD)and xanthine oxidase(XOD)levels were significantly increased with the increase of adeninein the culture medium(P<0.01).The levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and total antioxidant capacity(T-AOC)were increased significantly in each intervention group(P<0.01),while the level of nitric oxide(NO)was decreased significantly(P<0.01).The level of malondialdehyde(MDA)was increased at the beginning and then decreased.Compared with the control group,with the increase of UA level in Drosophila,the average life span of Drosophila in each intervention groups was significantly decreased(P<0.01),the amount of oviposition and climbing ability of female Drosophila in medium and high concentration intervention groups were significantly decreased(P<0.01),and climbing ability of male Drosophila in each intervention group was significantly decreased(P<0.01).Compared with the control group,the pupation rate and eclosion rate of Drosophila larvae in low concentration intervention group were significantly decreased(P<0.01).CONCLUSION:High concentration of UA inhibi
