摘　　要：目的建立超高效液相色谱-串联质谱(UPLC-MS/MS)法同时测定通窍耳聋丸(北柴胡、龙胆、黄芩等)中4种成分的含量。方法该药物甲醇提取液的分析采用Waters CORTECS T3色谱柱(2.1 mm×100 mm,2.7μm);流动相水-甲醇,梯度洗脱;体积流量0.4 mL/min;柱温35℃;电喷雾离子源,正离子检测;多反应监测模式。结果黄芩苷、龙胆苦苷、橙皮苷、柴胡皂苷A在各自范围内线性关系良好(R^(2)>0.9900),平均加样回收率97.45%~99.03%,RSD 0.3%~1.6%。结论该方法简便快速,灵敏度高,专属性强,稳定可靠,可用于通窍耳聋丸的质量控制。AIM To establish a ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)method for the simultaneous content determination of four constituents in Tongqiao Erlong Pills(Bupleuri Radix,Gentianae Radix et Rhizoma,Scutellariae Radix,etc.).METHODS The analysis of methanol extract of this drug was performed on a 35℃thermostatic Waters CORTECS T3 column(2.1 mm×100 mm,2.7μm),with the mobile phase comprising of water-methanol flowing at 0.4 mL/min in a gradient elution manner,and electron spray ionization source was adopted in positive ion detection with multiple reaction monitoring mode.RESULTS Baicalin,gentiopicroside,hemperidin and saikosaponin A showed good linear relationships within their own ranges(R^(2)>0.9900),whose average recoveries were 97.45%-99.03%with the RSDs of 0.3%-1.6%.CONCLUSION This simple,rapid,stab and reliable method demonstrates high sensitivity and strong specificity,which can be used for the quality control of Tongqiao Erlong Pills.
