川麦冬药材的特征图谱研究

摘　　要：目的:建立川麦冬药材的HPLC特征图谱,为科学评价和有效控制其质量提供可靠方法。方法::基于HPLC-ELSD法对20批次川麦冬药材进行特征图谱研究,采用ZORBAX SB-C_(18)色谱柱(4.6mm×250mm,5.0μm);以水(A)-乙腈(B)为流动相进行梯度洗脱;柱温30℃;流速1.0mL/min;进样量10μL,蒸发光散射检测器检测;漂移管温度:95℃,雾化器温度:90℃,气体流速:1.6L/min。结果:采用“中药色谱指纹图谱相似度评价系统”进行相似度评价,20批次川麦冬药材相似度均大于0.99,建立了川麦冬HPLC特征图谱,共有7个共有峰,并对其中4种甾体皂苷及高异黄酮类成分进行指认。结论:该研究所建立的川麦冬HPLC特征图谱的方法准确可行,可为川麦冬药材提供质量控制手段。Objective:Characteristic chromatograms of Ophiopogonis Radixfrom Sichuan were established by HPLC,and applied to provide a reliable method for scientific evaluation and effective quality control of this medicinal material.Methods:Characteristic chromatograms of 20batches of Ophiopogonis Radix from Sichuan were studied based on HPLC-ELSD.ZORBAX SB-C_(18)column(4.6mm×250mm,5.0μm)was performed for HPLC analysis.The mobile phase was water(A)-acetonitrile(B)with gradient elution,the column temperature was 30℃,the flow rate was 1.0mL/min and the injection volume was 10μL.The samples were detected by evaporative light scattering detector(ELSD),drift tube temperature at 95℃,the nebulizer temperature at 90℃and the gas flow rate was set at 1.6L/min.Results:The similarity of samples was evaluated by“Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine”and all over 0.99.HPLC characteristic chromatograms were established with 7common peaks,4steroidal saponins and homoisoflavonoids were identified.Conclusion:This method was accurate and viable,could be used for providing quality control of Ophiopogonis Radixfrom Sichuan.
