摘 要:目的分析比较2型糖尿病患者(T2D)与正常人群肠道菌群结构、丰度及多样性的差异。方法收集30例临床粪便样本(健康样本15例为研究组,2型糖尿病病例样本15例为对照组),应用Illumina MiSeq高通量测序技术进行Alpha、Beta多样性分析,比较研究组和对照组肠道菌群丰富度和多样性以及两组间的肠道菌群差异。采用Tax4Fun软件进行差异菌群功能预测。结果研究组和对照组OTU共有1565个,其中两组共有OTU为767个。与对照组比较,研究组肠道菌群α多样性下降;Unweighted Unifrac距离显著降低,差异有统计学意义(P<0.05),β多样性存在差异。在门水平上,研究组中厚壁菌门(Fimicutes)、变形杆菌门(Proteobacteria)的相对丰度明显高于对照组,而拟杆菌门(Bacteroidetes)、放线杆菌门(Actinobacteria)、疣微菌门(Verrucomicrobiota)相对丰度低于对照组。在属水平上,研究组克雷伯氏菌(Klebsiella)、普拉梭菌(Faecalibacterium)的相对丰度明显高于对照组,狄氏副拟杆菌(Parabacteroides)、普雷沃菌(Prevotella)、艾克曼菌(Akkermansia)、双歧杆菌(Bifidobacterium)的相对丰度明显低于对照组。线性判别效应(LEfSe)分析发现糖尿病可选择克雷伯菌属作为关键的生物标志物,对照组可选择双歧杆菌属、艾克曼菌属、普雷沃氏菌属、狄氏副拟杆菌属作为关键的生物标志物。通过差异菌群进行KEGG代谢通路分析发现组间25条KEGG代谢通路存在显著差异,研究组的能量代谢和糖代谢等代谢途径均有上调。结论研究组和对照组肠道菌群丰度上具有显著差异,可作为2型糖尿病诊断和治疗靶标的候选标志菌群。Objective To analyze and compare the differences in structure,abundance and diversity of intestinal flora between type 2 diabetic patients(T2D)and normal population.Methods 30 clinical stool samples(15 healthy samples and 15 samples from type 2 diabetes cases)were collected,and Alpha and Beta diversity analysis was performed by applying Illumina MiSeq high-throughput sequencing technology to compare the abundance and diversity of intestinal flora between the diabetic and thenormal groups and the differences in intestinal flora between the two groups.Differential flora function prediction was performed using Tax4Fun software.Results There were 1565 OTUs in the study and thecontrol groups,of which 767 OTUs were shared between the two groups.Compared with the control group,there was a decreasing inαdiversity of the intestinal flora in the study group;Unweighted Unifrac distance was significantly lower with statistically significant differences(P<0.05),and there were differences inβdiversity.At the phylum level,the relative abundance of Fimicutes and Proteobacteria in the study group was significantly higher than that of the control group,while the relative abundance of Bacteroidetes,Actinobacteria,and Verrucomicrobiota were lower than those of the normal group.At the genus level,the relative abundance of Klebsiella and Faecalibacterium was significantly higher in the study group than the control group,and the relative abundance of Parabacteroides,Prevotella,Akkermansia and Bifidobacterium was significantly lower than that of the control group.Linear discriminant effect(LEfSe)analysis revealed that Klebsiella spp.could be selected as the key biomarkers for diabetes,and Bifidobacterium spp.,Akkermansia spp.,Prevotella spp.and Dictyostelium parapsilosis spp.could be selected as the key biomarkers in the control group.KEGG metabolic pathway analysis by differential flora revealed the significant differences in 25 KEGG metabolic pathways between the groups,and metabolic pathways,such as energy metabolism and glucose me
