摘  要:目的研究阿纳其根醇提取物(EEAP)对咳嗽变异性哮喘大鼠肠道菌群的影响。方法采用卵清蛋白-费氏完全佐剂将6~8周龄雄性SD大鼠建立咳嗽变异性哮喘(Cough variant asthma,CVA)模型,随机分为3组,CVA模型组(等体积的生理盐水)、EEAP组(EEAP,640 mg/kg)、阳性药组(醋酸泼尼松,250 mg/kg),每组8只;取8只未造模的SD大鼠作为对照组(等体积的生理盐水)。EEAP和阳性药组大鼠给予药物连续干预,其余各组给予生理盐水,30 d后收集各组大鼠粪便。采用高通量测序技术分析各组大鼠肠道菌群,基于扩增子序列变异(Amplicon sequence variation,ASV)进行Alpha和Beta多样性分析EEAP对肠道细菌属水平的影响。结果高通量测序得到4771个ASV,Alpha多样性分析发现4组大鼠肠道菌群chao1、dominance、observed、pielou、shannon和simpson指标无明显变化,差异无统计学意义(P>0.05)。与对照组相比,CVA组大鼠肠道中Prevotellaceae_Ga6A1_group丰度明显升高,而Prevotella、Prevotellaceae_NK3B31_group和Prevotellaceae_UCG_001丰度明显降低,差异具有统计学意义(P<0.05);与CVA组相比,EEAP组和阳性药组大鼠肠道中Prevotella和Prevotellaceae_NK3B31_group丰度明显升高,而Prevotellaceae_Ga6A1_group丰度明显降低,差异有统计学意义(P<0.05)。结论EEAP对CVA的改善作用可能与调节肠道菌群结构有关,其机制需进一步研究。Objective To study the effect of ethanol extract of root of Anacyclus pyrethrum(EEAP)on the intestinal flora in rats with cough variant asthma.Methods Cough variant asthma(CVA)models were established in 6-8 week old male SD rats with ovalbumin Fisher complete adjuvantand were randomly divided into 3 groups:CVA model group(equal volume of normal saline),EEAP group(EEAP,640 mg/kg)and positive drug group(prednisone ace-tate,250 mg/kg),with 8 rats in each group;8 SD rats without model were taken as the control group(equal volume of normal saline).The rats in EEAP and positive drug groups were given continuous drug intervention,the rest groups were given normal saline,and the feces of rats in each group were collected 30 days later.High throughput sequencing technology was used to analyze the intestinal flora of the rats in each group.Alpha and Beta diversity analysis was conducted based on Amplicon sequence variation(ASV)to analyze the effect of EEAP on the level of intestinal bacteria.Results 4771 ASVs were obtained by high-throughput sequencing.Alpha diversity analysis showed that the 4 groups of rats had no significant changes in the indicators of chao1,dominance,observed,pielou,shannon and simpson,and the differences were not statistically significant(P>0.05).Compared with the control group,the abundance of Prevotellaceae_Ga6A1_group in the intestine of the rats in the CVA group were significantly increased,while the abundance of Prevotella,Prevotellaceae_NK3B31_groupand Prevotellaceae_UCG_001 were significantly decreased,and the difference were statistically significant(P<0.05).Compared with CVA group,the abundance of Prevotella and Prevotelaceae_NK3B31_group in the intestine of rats in EEAP group and positive drug group wereincreased significantly,while the abundance of Prevotelaceae_Ga6A1_group wasdecreased significantly(P<0.05).Conclusion The improvement effect of EEAP on CVA may be related to the regulation of intestinal flora structure,and the mechanism needs to be further studied.
