摘  要:【目的】对西南地区32个主推桑品种中α-生育酚含量进行测定分析,筛选出高α-生育酚含量材料,为高生育酚含量桑品种选育提供优异材料,奠定物质基础。【方法】利用HPLC法对桑叶及桑椹中α-生育酚含量进行系统测定。HPLC法采用C18色谱柱(250 mm×4.6 mm,5μm),流动相采用色谱纯甲醇(A)-超纯水(B)梯度洗脱,进样量10μL,流速1 mL/min,柱温20℃,走样时间为35 min,检测波长294 nm。采用SPSS 23.0统计软件对检测结果进行描述性统计、单因素ANOVA分析及Duncan多重比较、系统聚类分析。【结果】32个参试桑树叶片的α-生育酚含量介于13.44~64.14μg/g,平均含量为32.78μg/g;13个品种的桑椹α-生育酚含量介于0.48~13.1μg/g,平均含量仅为5.48μg/g,桑叶的α-生育酚平均含量为桑椹的5.98倍;叶片α-生育酚含量最高的品种是农桑2号,为64.14μg/g,最低的品种是湖桑14号,仅为13.44μg/g。从所属桑系角度看,鲁桑系桑树的叶片α-生育酚含量范围在12.91~64.52μg/g,均值为34.88μg/g,含量较高;山桑系和白桑系的α-生育酚含量较低,均值分别为30.58、31.51μg/g。聚类分析显示,32个叶用桑资源可聚类为3个大类,根据其α-生育酚含量,归纳为中等含量组、低含量组及较高含量组3类。【结论】鉴定出农桑2号、育2号、乐山红花桑、农桑8号、荷叶白等5个高α-生育酚含量品种,23个中等含量及4个低含量桑品种资源,为桑树资源的发掘利用及品质改良育种提供了理论依据和技术支撑。【Objective】The content ofα-tocopherol in 32 main mulberry varieties in Southwest China was determined and analyzed,and the materials with highα-tocopherol content were screened out,which provided excellent materials for the breeding of mulberry varieties with high tocopherol content and laid the material foundation.【Method】Theα-tocopherol content in mulberry leaves and mulberry fruits were determined by High Performance Liquid Chromatography(HPLC).HPLC was performed on C18 chromatographic column(250 mm×4.6 mm,5μm)with gradient elution of pure methanol(A)and ultrapure water(B)as mobile phase.The sample volume was 10μL,the flow rate was 1 mL/min,the column temperature was 20℃,the sample removal time was 35 min,and the detection wavelength was 294 nm.SPSS 23.0 statistical software was used for descriptive statistics,one-way ANOVA analysis,Duncan multiple comparison and systematic cluster analysis.【Result】Theα-tocopherol content in 32 mulberry leaves ranged from 13.44 to 64.14μg/g,with the average content of 32.78μg/g.Theα-tocopherol content in 13 mulberry fruits ranged from 0.48 to 13.1μg/g,the average content was only 5.48μg/g,The average ofα-tocopherol content in mulberry leaves was 5.98 times that in mulberry fruits.The variety with the highest content in leaves was‘Nongsang 2’,which was 64.14μg/g;The lowest variety was‘Husang 14’,only 13.44μg/g.From the perspective of mulberry family,the average content ofα-tocopherol in leaves of Morus multicaulis ranged from 12.91 to 64.52μg/g,with the average of 34.88μg/g,and the average content ofα-tocopherol in leaves of M.bombyilis and M.alba was 30.58,31.51μg/g,respectively.According to the content ofα-tocopherol,the 32 mulberry leaf resources could be classified into three categories:Medium content,low content and high content.【Conclusion】Five varieties with highα-tocopherol content,including Nongsang 2,Yu 2,Leshan Honghua sang,Nongsang 8 and Heye Bai were identified,and 23 varieties with medium content and 4 varieties with
