摘 要:目的以槲皮素(QT)为模型药,制备一种可再分散的槲皮素纳米晶自稳定Pickering乳液冻干粉(quercetin nanocrystals self-stabilized Pickering emulsion lyophilized powder,QT-NSSPE-LP)。方法采用高压均质法联合探头超声法制备QT-NSSPE,随后通过冻干技术实现其脱水固化得到QT-NSSPE-LP。以外观性状、复溶粒径和再分散指数(RDI)为评价指标,采用单因素试验筛选出QT-NSSPE-LP的优化处方,对优化条件得到的QT-NSSPE-LP进行表征和体外溶出评价。结果得到的QT-NSSPE-LP复溶粒径为(19.55±O.29)Mm,RDI为1.231±0.019;SEM和荧光显微镜结果表明冻干和再分散过程并未破坏QT-NSSPE-LP的形态结构。X-射线衍射结果表明QT-NSSPE-LP中QT的结晶度比原料药降低。体外溶出显示QT-NSSPE-LP的溶出度显著高于QT原料药。结论QT-NSSPE的固化可以通过冻干技术制备,能较好地保留Pickering乳液的形态结构和性质,可以为QT的进一步开发提供参考。Objective To develop a redispersible quercetin(QT)nanocrystalline self-stabilizing Pickering emulsion freeze-dried powder(QT-NSSPE-LP)with QT as a model drug.Methods QT-NSSPE was prepared using high-pressure homogenization and probe ultrasonic method before the freeze-drying method was employed to prepare QT-NSSPE-LP.With the appearance,reconstituted particle size,and redispersibility index(RDI)as evaluation indexes,the optimal prescription was screened out on a single factor test.QT-NSSPE-LP prepared under optimal conditions was characterized and evaluated in vitro.Results The redispersion size and RDI of the obtained QT-NSSPE-LP were(19.55±0.29)ym and 1.231±0.019,respectively.SEM and fluorescence microscopy results showed that the structure of QT-NSSPE-LP had not been destroyed in the process of freeze-drying and redispersion.X-ray diffraction showed that the crystallinity of QT in QT-NSSPE-LP was decreased compared with raw materials of QT.The result of the in vitro release experiment showed that QT-NSSPE-LP had a higher accumulative release rate than the raw materials of QT.Conclusion QT-NSSPE can be solidified using the freeze-drying technology,which can maintain the structure and property of Pickering emulsion.This study can provide reference for the further development of QT.
