摘  要:MiR-17-5p是在多种动物组织中广泛表达的一种miRNA,参与机体的性腺发育及繁殖等。本研究采用RT-qPCR方法检测miR-17-5p在60日龄尼罗罗非鱼(Oreochromis niloticus)幼鱼精巢和卵巢中的差异表达及其对雌二醇的应答反应特性,并对其交集靶基因进行GO富集分析和KEGG pathway分析。结果表明:miR-17-5p在尼罗罗非鱼幼鱼的精巢和卵巢中均有表达,卵巢中的表达量显著高于精巢中(P<0.01)。雌二醇用无水乙醇溶解后,再与芝麻油按照体积比1∶10稀释后,给体质量(43.1±0.2)g的尼罗罗非鱼腹腔注射浓度为10 mg/kg体质量。对照组注射无水乙醇与芝麻油(体积比1∶10)的混合液,2 d、7 d、14 d、21 d、28 d后取样。结果发现,雌二醇处理的尼罗罗非鱼精巢和卵巢中miR-17-5p的表达量显著降低(P<0.01)。GO富集分析表明,miR-17-5p的靶基因参与生长、发育、代谢和蛋白质修饰等相关过程,且多个靶基因富集到了与繁殖相关的经典WNT信号通路。KEGG pathway分析表明,miR-17-5p的多个靶基因富集于MAPK、TGF-beta、WNT和孕酮调节的卵母细胞成熟等通路。这些通路均与性腺发育和繁殖相关。本研究可为探究miR-17-5p在鱼类性别控制和性腺发育等过程的功能和分子机理提供基础数据。In this study,the expression differences of miR-17-5p as a kind of gene widely expressed in various animal tissues and involved in the gonadal development and reproduction were investigated in the testes and ovaries of 60 days old juvenile Nile tilapia(Oreochromis niloticus)by RT-qPCR,and then the response of the miRNA to estradiol in the testes and ovaries was also analyzed in the Nile tilapia with body weight of(43.1±0.2)g injected intraperitoneally with the mixture of estradiol dissolved in absolute ethanol with sesame oil in a volume ratio of 1∶10 at a dose of 10 mg/kg body weight.In the control group the fish was injected with a mixture of absolute ethanol and sesame oil(volume ratio 1∶10).After injection,gonads were obtained at 2 d,7 d,14 d,21 d,and 28 d.Finally,multiple softwares were used to predict the target genes of miR-17-5p and GO enrichment and KEGG pathway analysis were conducted on the intersection target genes.The result showed that miR-17-5p was expressed in the testes and ovaries of Nile tilapia,very sinificantly lower expression level of miR-17-5p in the testis than that in the ovary(P<0.01).The expression level of miR-17-5p in the testis and ovaries was significantly decreased in the Nile tilapia injected intraperitoneally with the mixture of estradiol.The GO enrichment analysis revealed that target genes of miR-17-5p were involved in growth,development,metabolism,protein modification and other related processes,and multiple target genes were enriched into the classical WNT signaling pathway related to reproduction.The KEGG pathway analysis showed that multiple target genes of miR-17-5p were enriched to the MAPK signaling pathway,TGF-beta signaling pathway,WNT signaling pathway and progesterone mediated oocyte maturation pathway,which were involved in reproduction and gonad development.The finding provides basis for exploring the function and molecular mechanism of miR-17-5p in the process of fish sex control and gonadal development.
