摘  要:目的研究左旋紫草素对HTR-8/Svneo细胞上皮间充质转化(epithelial-mesenchymal transition,EMT)的影响。方法将HTR-8/Svneo细胞用0,0.2,0.4,0.8和1.6μmol·L(-1)左旋紫草素干预6,12和24 h后,用CCK-8检测细胞活力,筛选出药物的最佳作用浓度和作用时间以进行后续实验。实验分为空白组、对照组和实验组。空白组不给予干预;对照组给予0.08 ng·L(-1)甲氨蝶呤干预,实验组给予0.8μmol·L(-1)左旋紫草素干预。用细胞划痕实验检测细胞迁移情况,用Western blot法检测EMT及p38 MAPK通路相关蛋白的表达水平。结果左旋紫草素的最佳作用浓度为0.8μmol·L(-1),最佳作用时间为24 h。实验组、对照组和空白组的迁移率分别为(18.39±1.25)%,(15.56±1.59)%和(29.58±2.56)%,E-cadherin蛋白相对表达水平分别为0.94±0.06,0.89±0.06和0.36±0.02,N-cadherin蛋白相对表达水平分别为0.87±0.08,0.82±0.06和1.31±0.12,Vimentin蛋白相对表达水平分别为0.89±0.09,0.86±0.07和1.19±0.09,p-p38蛋白相对表达水平分别为0.62±0.03,0.60±0.01和0.75±0.01,实验组的上述指标与空白组相比,差异均有统计学意义(P<0.05,P<0.01)。结论左旋紫草素可抑制HTR-8/Svneo细胞的增殖、迁移、侵袭能力,其作用机制可能与p38 MAPK信号通路有关。Objective To study the effect of L-shikonin on epithelial-mesenchymal transition(EMT)of HTR-8/Svneo cells.Methods After HTR-8/Svneo cells were treated with 0,0.2,0.4,0.8 and 1.6μmol·L(-1)L-shikonin for 6,12 and 24 h,the cell viability was detected by CCK-8,and the best drug was selected.Action concentration and action time for subsequent experiments.The experiment was divided into blank,control and experimental groups.Blank group was not given intervention.Control group was given 0.08 ng·L(-1)methotrexate.Experimental group was given 0.8μmol·L(-1)L-shikonin.Cell migration was detected by cell scratch assay,and the expression levels of EMT and p38 MAPK pathway-related proteins were detected by Western blot.Results The optimum concentration of L-shikonin was 0.8μmol·L(-1),and the optimum duration of action was 24 h.The migration rates of the experimental,control and blank groups were(18.39±1.25)%,(15.56±1.59)%and(29.58±2.56)%the relative expression levels of E-cadherin protein were 0.94±0.06,0.89±0.06 and 0.36±0.02;the relative expression levels of-cadherin protein were 0.87±0.08,0.82±0.06 and 1.31±0.12;the relative expression levels of Vimentin protein were 0.89±0.09,0.86±0.07 and 1.19±0.09;the relative expression levels of p-p38 protein were 0.62±0.03,0.60±0.01 and 0.75±0.01;the above indexes in the experimental group were significantly different from those in the blank group(P<0.05,P<0.01).Conclusion L-shikonin can inhibit the proliferation,migration and invasion of HTR-8/Svneo cells,and its mechanism may be related to the p38 MAPK signaling pathway.
