摘  要:炎症是生物体内与许多疾病相关的重要保护反应,过氧亚硝酸根(ONOO-)参与炎症过程,与炎症有着密切联系。采用6-羟基-2,3,4,4a-四氢-1H-氧杂蒽-1-酮(GCTPOC)作为双光子荧光团(双光子荧光活性截面δΦ=262 GM(1 GM=1×10^(-50)cm^(4)·s·photons^(-1)·molecule^(-1)),激发波长λ_(ex)=860 nm)设计并合成一种用于检测过氧亚硝酸根的双光子荧光探针GCTONOO。该探针具有良好的荧光响应,检出限低(13.5 nmol/L)、Stokes位移大(135 nm),可特异性识别ONOO^(-)。细胞急性炎症模型实验结果表明,该探针可实现细胞炎症过程中ONOO^(-)的荧光成像分析。Inflammation is an important protection reaction in living organisms associated with many diseases. Peroxynitrite(ONOO-) is involved in the inflammatory processes and has an important relationship with inflammation. Herein, a two-photon fluorescence probe GCT-ONOO for detecting peroxynitrite was designed and synthesized using 6-hydroxy-2,3,4, 4a-tetrahydro-1H-oxanthracene-1-one(GCTPOC) as the two-photon fluorophore(two-photon fluorescence activity cross section δΦ=262 GM(1 GM=1×10^(-50)cm^(4)·s·photons^(-1)·molecule^(-1)),excitation wavelength λ_(ex)=860 nm). The probe can specifically recognize ONOO^(-) with good fluorescence response, low detection limit(13.5 nmol/L) and large Stokes displacement(135 nm). The results of cell experiment in acute inflammation model showed that the probe can achieve highly sensitive imaging analysis for peroxynitrite during cellular inflammation.
