摘  要:目的:探究黄芩茎叶对2%角叉菜胶致急性胸膜炎大鼠模型的抗炎作用,以期探索其作用机制及对TRPV1信号蛋白的影响。方法:将60只健康SPF级雄性大鼠随机分为:正常对照组、模型对照组、地塞米松片5 mg/kg组、黄芩茎叶3、6、12 g/kg组。各组灌胃给予相应药物或生理盐水,连续7 d,1次/d,末次给药2 h后用乙醚轻微麻醉,除正常对照组外,其余大鼠于右胸腔第7肋间向内注射2%角叉菜胶建立急性胸膜炎模型,6 h后麻醉,颈椎脱臼处死。测定胸腔渗出液中白细胞数量和蛋白含量;酶联免疫吸附法(ELISA)检测血清中前列腺素E2(PGE)、白细胞介素-6(IL-6)和肿瘤坏死因子(TNF-α)含量;HE染色检查肺组织病理变化及损伤评分;实时荧光定量(RT-PCR)分析肺组织中Trpv1 mRNA表达;蛋白免疫印迹法(Western blot)检测肺组织中TRPV1蛋白的表达。结果:与正常对照组相比,模型对照组胸腔渗出液中白细胞数和蛋白含量显著升高(P<0.01),血清中的PGE、IL-6和TNF-α炎性因子含量显著增加,肺泡组织结构破坏,可见大量中性粒细胞,肺泡壁增厚明显,TRPV1蛋白表达显著上调(P<0.01);与模型对照组相比,黄芩茎叶6、12 g/kg组及地塞米松片5 mg/kg组胸腔渗出液中白细胞数和蛋白含量明显降低,肺组织病变明显改善,血清中PGE、IL-6和TNF-α炎性因子含量明显减少(P<0.05或P<0.01),TRPV1蛋白表达明显下调(P<0.05或P<0.01)。结论:黄芩茎叶对2%角叉菜胶致大鼠急性胸膜炎具有抗炎作用,作用机制可能与降低机体炎性因子产生和下调TRPV1信号有关。Objective:To investigate the anti-inflammatory effects of Scutellaria baicalensis Georgi stems and leaves(SBSL) on 2% carrageenan-induced acute pleurisy in rats to explore the underlying mechanism of action and the effect on TRPV1 signaling protein. Methods:Sixty healthy male rats of SPF grade were randomly divided into a normal control group, a model control group, a dexamethasone tablet group(5 mg/kg),and high-(12 g/kg),medium-(6 g/kg),and low-dose(3 g/kg) SBSL groups. Drug intervention was carried out by gavage for 7 days, once per day. Two hours after the last administration, rats were lightly anesthetized with ether. The acute pleurisy model was induced by internal injection of 2% carrageenan into the 7 th intercostal space of the right thoracic cavity in rats except for those in the normal control group. The rats were anesthetized 6 h later and sacrificed by cervical dislocation. The white blood cell count and protein content in the pleural exudate were measured. The serum levels of prostaglandin E(PGE),interleukin-6(IL-6),and tumor necrosis factor(TNF)-α were detected by enzyme-linked immunosorbent assay(ELISA). The pathological changes and injury scores of lung tissues were checked by hematoxylin-eosin(HE) staining. The mRNA and protein expression of TRPV1 in lung tissues was analyzed by real-time fluorescence-based quantitative polymerase chain reaction(RT-PCR) and Western blot, respectively. Results:Compared with the normal control group, the model control group showed increased white blood cell count and protein content in the pleural exudate(P<0.01),increased serum levels of PGE,IL-6,and TNF-α,damaged alveolar tissue structure, massive neutrophile granulocytes, thickening of alveolar wall, and elevated protein expression of TRPV1(P<0.01). Compared with the model control group, the groups with drug intervention showed decreased white blood cell count and protein content in the pleural exudate, relieved lesions in lung tissue, reduced serum levels of PGE,IL-6,and TNF-α(P<0.05 or P<0.01),and dwindled
