摘 要:目的:研究红景天苷在Raw 264.7细胞炎症模型中对线粒体功能和炎症损伤的影响。方法:MTS方法检测红景天苷(1、10、50、100μg/mL)对Raw264.7细胞活力的影响,筛选低、中、高浓度;LPS(1μg/mL)构建Raw 264.7细胞炎症模型。将细胞分为空白对照组、模型组、红景天苷组、甲氨蝶呤组、AG490(JAK2/STAT3抑制剂)组、红景天苷+Colivelin(JAK2/STAT3激活剂)组;ELISA法检测IL-6、IL-1β、TNF-α、ROS水平,Western Blot检测IL-6、IL-1β、TNF-α、p-JAK2、p-STAT3、JAK2、STAT3蛋白表达;试剂盒检测细胞内ATP水平。结果:红景天苷低、中、高浓度分别为1、10、50μg/mL,对Raw 264.7细胞活力均无明显影响。与空白对照组比较,模型组IL-6、IL-1β、TNF-α分泌量及ROS、ATP水平和IL-6、IL-1β、TNF-α、p-JAK2、p-STAT3蛋白表达显著升高(P<0.05)。与模型组比较,红景天苷10、50μg/mL组IL-6、IL-1β、TNF-α分泌量及IL-6、IL-1β、TNF-α蛋白表达和ROS、ATP水平显著降低,红景天苷50μg/mL组p-JAK2、p-STAT3蛋白表达显著降低(P<0.05)。AG490与红景天苷作用一致,而与红景天苷组比较,红景天苷+Colivelin组ROS、ATP水平及IL-6、IL-1β、TNF-α分泌量显著升高(P<0.05)。结论:红景天苷通过抑制JAK2/STAT3通路调控线粒体功能,抵抗LPS诱导的Raw 264.7细胞炎性损伤。Objective: To study the effects of salidroside on mitochondrial function and inflammatory injury in Raw 264.7 cells inflammatory model.Methods: MTS method was used to detect the effects of salidroside(1,10,50,100 μg/mL)on Raw 264.7 cells activity, and low, medium and high concentrations were screened.1 μg/mL LPS was used to construct the Raw 264.7 cells inflammatory model, and the cells were divided into blank control group, model group, salidroside group, methotrexate group, AG490(JAK2/STAT3 inhibitor) group, salidroside with Colivelin(JAK2/STAT3 activator)group.The levels of IL-6,IL-1β,TNF-α,ROS were detected by ELISA.The expressions of IL-6,IL-1β,TNF-α,p-JAK2,p-STAT3,JAK2,STAT3 proteins were detected by Western Blot.The kit was used to detect intracellular ATP level.Results: The low, medium, high concentrations of salidroside were 1,10,50 μg/mL,respectively, which had no significant effect on the activity of Raw 264.7 cells.Compared with blank control group, the secretions of IL-6,IL-1β and TNF-α,the levels of ROS and ATP,and the protein expressions of IL-6,IL-1β,TNF-α,p-JAK2,p-STAT3 in model group were significantly increased(P<0.05).Compared with model group, the secretions of IL-6,IL-1β and TNF-α,the protein expressions of IL-6,IL-1β,TNF-α and the levels of ROS,ATP in salidroside 10 and 50 μg/mL groups were significantly decreased, while the protein expressions of p-JAK2 and p-STAT3 in salidroside 50 μg/mL group were significantly decreased(P<0.05).AG490 had the same effect as salidroside, while the levels of ROS and ATP and the secretions of IL-6,IL-1β and TNF-α in salidroside with Colivelin group were significantly increased compared with salidroside group(P<0.05).Conclusion: Salidroside regulates mitochondrial function against LPS-induced inflammatory injury of Raw 264.7 cells by inhibiting JAK2/STAT3 pathway.
