摘  要:目的探讨微塑料蓄积与血脂异常间的关系。方法将12只C57BL/6J小鼠分为空白组、正常组、高脂组、血脂康组,9只ApoE^(-/-)小鼠分为正常组、高脂组、血脂康组,其中,C57BL/6J小鼠和ApoE^(-/-)小鼠的高脂组、血脂康组给予高脂饲料,其他各组给予普通饲料。实验时,除空白组,其他各组小鼠灌胃含有绿色荧光的微塑料,全自动生化分析仪检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量,小动物活体光学成像系统(in vivo imaging system,IVIS)检测组织器官中微塑料荧光量。结果(1)微塑料灌胃2 h后,C57和ApoE^(-/-)小鼠均表现为高脂状态下微塑料的荧光强度增高,血脂康降脂治疗后荧光强度降低;(2)ApoE^(-/-)小鼠在正常和高脂状态下的各组织器官均表现为微塑料荧光强度高于C57小鼠;(3)C57和ApoE^(-/-)小鼠的血脂康组体内附睾、脑、内脏脂肪、皮下脂肪中的微塑料荧光明显降低,且ApoE^(-/-)小鼠体内微塑料荧光强度的降幅更大,但大多数组织器官中仍表现为ApoE^(-/-)小鼠体内微塑料荧光强度高于C57小鼠,与两种小鼠的血脂指标趋势相同;(4)相关性分析显示,组织器官中的微塑料荧光量基本呈现与TC、TG、LDL-C水平呈正相关,与HDL-C水平呈负相关的趋势,其中,肝脏中微塑料荧光量与TC、LDL-C相关性强,附睾中微塑料荧光量与HDL-C相关性强,脑中微塑料荧光量与TG、HDL-C、LDL-C相关性强,内脏脂肪中微塑料荧光量与TC、TG、HDL-C和LDL-C相关性均较强,皮下脂肪和棕色脂肪中微塑料荧光量与TC、TG和LDL-C相关性强。结论微塑料蓄积与TC、TG、HDL-C、LDL-C水平密切相关,健脾祛痰法降脂治疗后微塑料在机体内的蓄积量明显降低。Objective To investigate the relationship between accumulation of microplastics and abnormal blood lipids.Methods Twelve C57BL/6J mice were divided into blank group,normal group,high-fat group,and Xuezhikang(血脂康)group.9 ApoE^(-/-)mice were divided into normal group,highfat group,and Xuezhikang group.Among them,the high-fat group and Xuezhikang group of C57BL/6J mice and ApoE^(-/-)mice were given high-fat diet,and the other groups were given normal diet.During the experiment,except for the blank group,mice in the other groups contained green fluorescent microplastics,and the automatic biochemical analyzer detected serum total cholesterol(TC),triglycerides(TG),and high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C)content,small animal in vivo imaging system(IVIS)to detect the amount of microplastic fluorescence in tissues and organs.Results(1)After 2 hours of intragastric administration of microplastics,both C57 and ApoE^(-/-)mice showed that the fluorescence intensity of microplastics increased in the high-fat state,and the fluorescence intensity of Xuezhikang decreased after lipid-lowering treatment;(2)ApoE^(-/-)all tissues and organs of mice under normal and high-fat state showed that the fluorescence intensity of microplastics was higher than that of C57 mice;(3)The fluorescence of microplastics of epididymis,brain,visceral fat and subcutaneous fat in Xuezhikang group of C57 and ApoE^(-/-)mice were significantly reduced,and the fluorescence intensity of microplastics in ApoE^(-/-)mice had a greater decrease,but most tissues and organs still showed that the fluorescence intensity of microplastics in ApoE^(-/-)mice was higher than C57 mice with the same trend of blood lipid indexes as the two kinds of mice;(4)Correlation analysis showed that the amount of microplastic fluorescence in tissues and organs was basically positively correlated with TC,TG,LDL-C,and HDL-C levels.There was a negative correlation trend.Among them,the fluorescence of microplastics in the liver was st
