一例牛丘疹性口炎的诊断及病原序列分析

摘  要:为查明某奶牛场犊牛群出现口腔黏膜糜烂症状的病因,无菌采集10份发病犊牛口腔黏膜糜烂组织样本,对引起牛口腔黏膜糜烂的常见病原,通过PCR检测和基因测序进行病原鉴定和序列分析。结果显示:10份样本中检出6份牛丘疹性口炎病毒(BPSV)阳性,其他病原未检出;从6份BPSV阳性样本中,克隆获得6条部分B2L基因序列;经系统发育分析,6条B2L基因序列聚为两个不同的分支,并且每个分支内包含3条扩增获得的序列。结果表明,该奶牛场犊牛暴发的以口腔黏膜糜烂为特征的疾病为BPSV感染,而且牛场可能存在两种不同毒株流行。本研究丰富了国内BPSV毒株资料,为后续BPSV分子特征研究奠定了基础。In order to find out the causes leading to the symptoms of oral mucosa erosion in calves in a dairy farm,10samples were aseptically collected for pathogen identification and sequence analysis by PCR assay and gene sequencing.The results showed that 6 out of 10 samples were positive for bovine papular stomatitis virus(BPSV),and no any other pathogen was detected;6 partial B2L genes were cloned from the positive samples;it was showed that,by phylogenetic analysis,the gene sequences clustered into two different branches,in each of which three amplified sequences were contained. In conclusion,the disease characterized by oral mucosa erosion in the calves was caused by BPSV infection,and two different virus strains might be prevalent in the farm. By the study,the data of domestic BPSV strains was enriched,which laid a foundation for subsequent studies on molecular characteristics of BPSV.
