摘  要:绵羊肺炎支原体(Mycoplasma ovipneumoniae,Mo)可引起山羊、绵羊发生支原体性肺炎,分析绵羊肺炎支原体感染对贵州不同品种山羊Toll样受体(TLRs)基因转录水平的影响。应用Taq Man RT-qPCR对Mo人工感染的贵州黑山羊、贵州白山羊、黔北麻羊和波尔山羊的肺脏和血液样本TLRs基因转录水平进行定量检测与分析。结果显示,不同品种山羊感染Mo后,肺脏样本中贵州黑山羊和贵州白山羊TLR 1/2/4/6/8/9/10基因、黔北麻羊和波尔山羊TLR 4/6/7/8/10基因转录水平均极显著上调(P<0.01),血液样本中贵州黑山羊TLR 2/4/6/7/8/9基因、贵州白山羊TLR 2/4/6/7/8基因、黔北麻羊TLR 4/6/7/8/9基因、波尔山羊TLR 4/6/8/10基因转录水平均极显著上调(P<0.01);且发现不同品种山羊间TLRs基因的转录水平也存在显著差异,最先出现死亡的贵州白山羊、贵州黑山羊肺脏样本中TLR 1/6/8/10基因转录水平上调倍数显著高于其余品种山羊(P<0.05),而TLR 2/4/5/7/9基因转录水平上调倍数显著低于黔北麻羊和波尔山羊;贵州白山羊、贵州黑山羊血液样本中TLR 1/4/6/7/8基因转录水平高于黔北麻羊和波尔山羊,而TLR 2/5基因转录水平极显著低于黔北麻羊和波尔山羊(P<0.01),这种差异表达可能是不同品种山羊对Mo易感性不同的原因。结果表明,不同品种山羊对Mo感染的表现与TLRs基因转录水平有关,为研究Mo感染对不同品种山羊的致病机制提供科学参考。Mycoplasma ovipneumoniae(Mo)can cause mycoplasma pneumonia in goats and sheep.To analyze the effect of Mo infection on Toll-like receptors(TLRs)gene transcription levels in different breeds of goats in Guizhou can provide basic research data for analyzing the difference of Mo infection in different breeds of goats in Guizhou.Taq Man RT-qPCR was used to quantitatively detect and analyze the transcription levels of TLRs genes in lung and blood samples of Guizhou black goats,Guizhou white goats,Qianbei Ma goats and Boer goats artificially infected with Mo.The results showed that the transcription levels of TLR 1/2/4/6/8/9/10 gene in Guizhou black goats and Guizhou white goats,TLR 4/6/7/8/10 gene in Qianbei Ma goats and Boer goats were significantly up-regulated in lung samples after Mo infection(P<0.01).The transcription levels of TLR 2/4/6/7/8/9 gene in Guizhou black goats,TLR 2/4/6/7/8/8 gene in Guizhou white goats,TLR 4/6/7/8/9 gene in Qianbei Ma goats and TLR 4/6/8/10 gene in Boer goats were significantly up-regulated in blood samples(P<0.01).It was also found that there were significant differences in the transcription level of TLRs gene among different breeds of goats.The transcription level of TLR 1/6/8/10 gene in the lung samples of Guizhou white goats and Guizhou black goats with the first death was significantly higher than that of their breeds(P<0.05),while the transcription level of TLR 2/4/5/7/9 gene was significantly lower than that of Qianbei Ma goats and Boer goats.The transcriptions level of TLR 1/4/6/7/8 genes in blood samples of Guizhou white goats and Guizhou black goats was higher than that of Qianbei Ma goats and Boer goats,while TLR 2/5 gene was significantly lower than that of Qianbei Ma goats and Boer goats(P<0.01).This differential expression may be the reason for the different susceptibility of different goats breeds to Mo.The results showed that the expression of Mo infection in different breeds of goats in Guizhou was related to the transcription level of TLRs genes,which provided scient
