摘 要:为调查青岛地区鸡源大肠埃希氏菌的流行性及耐药情况,从青岛部分规模化养殖场随机采集185份鸡泄殖腔拭子,通过选择性培养基和16S rRNA进行大肠埃希氏菌分离纯化和菌属鉴定,采用琼脂稀释法对11种抗菌药物进行药敏试验并应用PCR检测碳青霉烯类、β-内酰胺类、黏菌素类和四环素类耐药基因。结果显示共分离到173株大肠埃希氏菌,分离率为93.50%。药敏试验结果表明,分离菌株对阿莫西林、头孢氨苄、四环素等8种抗菌药物耐药率为61.27%~100.00%,对美罗培南、亚胺培南、替加环素耐药率为0.58%~30.06%。分离菌株均表现为多重耐药(3耐~10耐),以7耐~8耐为主。耐药基因检测显示,bla_(CTX-M)和tet A检出率为95.95%和90.75%,其他耐药基因的检出率为3.47%~37.90%。结果表明,山东省青岛地区鸡源大肠埃希氏菌耐药率较高,多重耐药情况严重,给食品安全及公共健康造成了潜在威胁。In order to investigate the prevalence and drug resistance of E.coli from chickens in Qingdao,185 cloaca swabs were randomly collected from several large-scale breeding farms in Qingdao.The agar dilution method was used to test the susceptibility of 11 kinds of antibacterial drugs and the PCR was used to detect the resistance genes of carbapenems,β-lactams,quinolones,colistins and tetracyclines.The results showed that a total of 173 strains of Escherichia coli were isolated,with an isolation rate of 93.50%.The antimicrobial susceptibility test showed that the isolates to 8 antibiotics including amoxicillin,cephalexin,and tetracycline were 61.27%-100.00%;and the resistance rates to meropenem,imipenem and tigecycline were 0.58%-30.06%.In addition,all the isolates showed multi-drug resistance(resistance to 3-10 antibiotics),in which multi-drug resistance to 7-8 antibiotics was the predominant.The detection of resistance genes showed that the rates of bla_(CTX-M) and tet A were high at 95.95%and 90.75%,respectively;while the other gene resistance rates were 3.47%-37.90%.The results showed that the antibiotic-resistant rate of chicken-derived E.coli in Qingdao of Shandong province is relatively high,the situation of multi-drug resistance is serious,which poses a potential threat to food safety and public health.
