加味玉屏风及其多糖对小鼠免疫功能的影响

摘　　要：为研究加味玉屏风及其多糖对环磷酰胺诱导的免疫抑制小鼠免疫功能的影响,采用加味玉屏风和加味玉屏风多糖给药8 d,在第5~第7天腹腔注射环磷酰胺建立免疫抑制小鼠模型,末次给药24 h后,检测小鼠血常规、胸腺和脾脏指数,观察脾脏、胸腺组织学变化;采用流式细胞术检测脾脏中CD3^(+)和CD4^(+)T细胞数量比例、CD4^(+)/CD8^(+)T细胞比值;采用ELISA检测血清中sIgA、IgG、IgM含量。结果显示,加味玉屏风及其多糖均能明显提高免疫抑制小鼠的外周血白细胞数量、胸腺及脾脏指数、CD3^(+)和CD4^(+)T细胞数量比例、CD4^(+)/CD8^(+)T细胞比值,改善脾脏、胸腺病理组织损伤,回升血清中sIgA、IgG与IgM含量,且加味玉屏风多糖的效果优于加味玉屏风。表明加味玉屏风及其多糖可增强免疫抑制小鼠的免疫功能。To study the effects of Jiawei Yupingfeng and its polysaccharides on immune function of immunosuppressive mice induced by cyclophosphamide,Jiawei Yupingfeng and Jiawei Yupingfeng polysaccharides were administered for 8 days,the immunosuppressive mouse model was established by intraperitoneal injection of cyclophosphamide on the 5th-7th day,24 hours after the last administration,the blood routine,thymus and spleen indexes were measured,and the histological changes of spleen and thymus were observed.The quantity proportion of CD3+and CD4+T cells and the ratio of CD4+/CD8+of T cells in spleen were detected by flow cytometry,and the contents of sIgA,IgG and IgM in serum were detected by ELISA.The results showed that Jiawei Yupingfeng and its polysaccharides could significantly increase the number of peripheral blood leukocytes,thymus and spleen indexes,the quantity proportion of CD3+and CD4+T cells,and the ratio of CD4+/CD8+T cells,improve the pathological tissue damage of spleen and thymus,and increase the contents of sIgA,IgG and IgM in serum.The effect of Jiawei Yupingfeng polysaccharides is better than that of Jiawei Yupingfeng.The results indicates that Jiawei Yupingfeng and its polysaccharides can enhance the immune function of immunosuppressive mice.
