摘 要:初步探讨了淫羊藿多糖和板蓝根多糖的制备和理化性质,从而为进一步应用提供参考。用水煎醇沉法提取粗淫羊藿多糖(EPSg)和粗板蓝根多糖(IRPSg),经Sevag法除蛋白,DEAE-52柱层析分离,并对纯淫羊藿多糖EPS1和纯板蓝根多糖IRPS1进行沉淀溶解和红外光谱鉴定。结果显示,EPSg和IRPSg的提取率分别为4.24%和5.24%;经柱层析分离,分别得到2个纯淫羊藿多糖(EPS1和EPS2)和2个纯板蓝根多糖(IRPS1和IRPS2),EPS1的糖含量(53.1%)高于EPS2(37.34%),EPS1收集液用50 mL/L盐酸滴定产生的颗粒状沉淀用20 mg/mL NaOH溶液回滴不能溶解,IRPS1的糖含量(97.92%)高于IRPS2(30.85%),IRPS1收集液用50 mL/L盐酸滴定产生的絮状沉淀用20 mg/mL NaOH溶液回滴可以溶解;红外光谱显示,EPS1有酸性多糖的特征光谱,IRPS1无糖醛酸。结果表明,淫羊藿多糖与板蓝根多糖的组成和理化性质不完全相同。In this paper,the preparation and physicochemical properties of polysaccharides from Epimedium and Isatidis root were discussed,which could provide reference for further study and application.Gross Epimedium polysaccharide(EPSg)and gross Isatis root polysaccharide(IRPSg)were extracted by water decocting and alcohol precipitation method.The proteins were removed by Sevag means and separated by DEAE-52 column chromatography.EPS1 and IRPS1 were determined by precipitation and infrared spectra.The results showed that the extraction rates of EPSt and IRPSt were 4.24%and 5.24%,respectively.Two pure Epimedium polysaccharides(EPS1 and EPS2)and two pure Isatis root polysaccharides(IRPS1 and IRPS2)were obtained by column chromatography.The sugar content of EPS1(53.1%)was higher than that of EPS2(37.34%),the granular precipitation of EPS1 collected solution titrated with 50 mL/L hydrochloric acid could not be dissolved by 20 mg/mL NaOH solution.Sugar content of IRPS1(97.92%)was higher than that of IRPS2(30.85%),the flocculent precipitation of IRPS1 collected solution titrated with 50 mL/L hydrochloric acid could be dissolved by 20 mg/mL NaOH solution;Infrared spectra showed that EPS1 had the characteristic spectrum of acidic polysaccharide,and IRPS1 had no uronic acid.The results indicated that the compositions and physicochemical properties of polysaccharides from Epimedium and Isatis root were not identical.
