摘 要:为建立蕲蛇及其混伪品的生物信息学鉴别方法,采用随机扩增DNA多态性技术(RAPD)扩增蕲蛇线粒体细胞色素b(Cyt b),将扩增产物应用高效毛细管电泳技术(HPCE)和紫外线检测技术进行检测,对影响RAPD和HPCE的因素进行优化,建立蕲蛇的HPCE-RAPD指纹图谱,并对所得指纹图谱进行相似性分析发现,蕲蛇正品及伪品的指纹图谱相似性存在显著性差异。故本方法不仅适用于蕲蛇及其伪品的快速鉴别,再进一步研究的基础上也可广泛应用于其他易混中药的快速鉴别。In order to establish a bioinformatics identification method for Agkistrodon acutus and its adulterants,the mitochondrial cytochrome b(Cyt b)of Agkistrodon acutus was amplified by randomly amplified DNA polymorphism(RAPD).The amplified products were detected by high performance capillary electrophoresis(HPCE)and ultraviolet detection technology.The factors affecting RAPD and HPCE were optimized,the HPCE-RAPD fingerprint of Agkistrodon acutus was established.Meanwhile the similarity of the obtained fingerprint was analyzed.It was found that there were significant differences in the similarity of the fingerprints between the authentic and counterfeit Agkistrodon acutus.Therefore,this method is not only suitable for the rapid identification of Agkistrodon acutus and its counterfeits,but also can be widely used for the rapid identification of other easily mixed Chinese medicines on the basis of further research.
