摘  要:目的 研究不同浓度钙卫蛋白在滋养细胞凋亡过程中的影响,进而分析其在妊娠高血压疾病发病机理中的作用。方法 应用MTT法测定滴加钙卫蛋白后滋养细胞的吸光度值,以表达增殖活性,观察滋养细胞在透射电镜下形态结构的改变。结果 24 h和48 h的钙卫蛋白分别作用后,滋养细胞增殖活性随钙卫蛋白浓度增加而降低(r=-0.735;-0.750,P<0.01)。通过透射电镜观察,滋养细胞经钙卫蛋白处理后呈凋亡特征。结论 钙卫蛋白对滋养细胞有诱导凋亡作用,且凋亡率随药物浓度增加而升高。Objective To investigate the effect of different concentrations of calprotectin on apoptosis of trophoblast cells, and elucidate the role of calprotectin in the pathogenesis of hypertensive disorder complicating pregnancy.Methods Human calprotectin was treated on trophoblast cells of early pregnancy in primary culture, the effect of calprotectin on the proliferative activity of trophoblast cells was measured by MTT method, the ultrastructural changes of trophoblast cells after calprotectin action were observed by transmission electron microscopy. Results A negative correlation was observed between the drug concentration and the proliferative activity of trophoblast cells after 24 and 48 hours of calprotectin action(r=-0.735;-0.750, P<0.01). Under transmission electron microscopy, the trophoblast cells showed apoptotic features after treatment with calprotectin. Conclusion Calprotectin can induce apoptosis in trophoblast cells, and the apoptosis rate of trophoblast cells was positively correlated with the concentration of calprotectin.
