摘  要:目的 探讨右美托咪定对丙泊酚致新生大鼠海马神经元凋亡与远期空间学习记忆能力的影响。方法 将72只7 d龄SD大鼠利用随机数字表法随机分6组(n=12),分别为右美托咪定组、丙泊酚组、25μg/kg右美托咪定+丙泊酚组、50μg/kg右美托咪定+丙泊酚组、75μg/kg右美托咪定+丙泊酚组。术后1 d进行海马组织Western blot检测、炎性因子检测与免疫组化染色,术后60 d进行Morris水迷宫实验。结果 与对照组比较,丙泊酚组Bcl-2蛋白相对表达量减少,Bax、Caspase-3蛋白相对表达量增加;与丙泊酚组比较,50μg/kg右美托咪定+丙泊酚组、75μg/kg右美托咪定+丙泊酚组Bcl-2蛋白相对表达量增加,Bax、Caspase-3蛋白相对表达量减少。与对照组比较,丙泊酚组海马组织内的TNF-α、IL-1β、IL-6浓度明显升高;与丙泊酚组比较,50μg/kg右美托咪定+丙泊酚组、75μg/kg右美托咪定+丙泊酚组海马组织内的TNF-α、IL-1β、IL-6浓度降低。与对照组比较,丙泊酚组海马内的c-fos表达增加;与丙泊酚组比较,50μg/kg右美托咪定+丙泊酚组、75μg/kg右美托咪定+丙泊酚组海马内的c-fos表达减少。与对照组比较,丙泊酚组逃避潜伏期明显延长,穿越平台次数明显减少;与P组比较,50μg/kg右美托咪定+丙泊酚组、75μg/kg右美托咪定+丙泊酚组逃避潜伏期明显缩短,穿越平台次数明显增加。结论 50μg/kg、75μg/kg的右美托咪定可降低丙泊酚造成的神经损害与远期空间学习记忆能力障碍,其作用机制可能与抑制神经元凋亡、减轻神经炎症及抑制c-fos蛋白表达有关。Objective To investigate the effect of dexmedetomidine on the apoptosis of hippocampal neurons and long-term spatial learning and memory ability in neonatal rats induced by propofol. Methods 72 7-day-old SD rats were randomly divided into 6 groups(n=12) by random number table method: Dexmedetomidine group, propofol group, 25 μg/kg dexmedetomidine+propofol group, 50 μg/kg dexmedetomidine+propofol group and 75 μg/kg dexmedetomidine+propofol group. Western blot detection, inflammatory factor detection and immunohistochemical staining of hippocampal tissue were performed 1 day after operation, and Morris water maze test was performed 60 days after operation. Results Compared with the control group, the relative expression of Bcl-2 protein decreased and the relative expression of Bax and caspase-3 protein increased in propofol group. Compared with propofol group, the relative expression of Bcl-2 protein increased and the relative expression of Bax and caspase-3 protein decreased in 50 μg/kg dexmedetomidine+propofol group and 75 μg/kg dexmedetomidine+propofol group. Compared with the control group, TNF in hippocampus of propofol group-α, IL-1 β, the concentration of IL-6 increased significantly. Compared with propofol group, TNF in hippocampus of 50 μg/kg dexmedetomidine+propofol group and 75 μg/kg dexmedetomidine+propofol group-α, IL-1 β, IL-6 concentration decreased. Compared with the control group, the expression of c-fos in hippocampus of propofol group increased. Compared with propofol group, the expression of c-fos in hippocampus of 50 μg/kg dexmedetomidine+propofol group and 75 μg/kg dexmedetomidine+propofol group decreased. Compared with the control group, the escape latency was significantly prolonged and the number of crossing the platform was significantly reduced in the propofol group. Compared with group P, 50 μg/kg dexmedetomidine+propofol group and 75 μg/kg dexmedetomidine+propofol group significantly shortened the escape latency and significantly increased the number of crossing the platf
