摘 要:类受体胞质激酶(receptor-like cytoplasmic kinases,RLCKs)是一类缺乏跨膜结构域和胞外结构域,但含胞内激酶域的类受体激酶。RLCKs在植物生长发育与逆境胁迫应答生理过程中扮演重要的角色,目前番茄中仅少数几个RLCKs的功能被报道。为探究番茄RLCK家族成员的进化和功能,本研究通过生物信息分析鉴定到121个番茄RLCKs,归入17个亚家族。分析表明,番茄RLCK家族成员在DNA水平和蛋白水平上分别具有多样性和保守性,且在番茄12条染色体上呈不均匀分布,片段复制驱使了番茄RLCK家族成员的扩张。组织表达分析显示,番茄RLCK呈现不同的组织表达模式,在番茄各发育阶段均有番茄RLCK的表达。微阵列数据分析显示,数个番茄RLCKs分别在生物逆境因子真菌诱导子EIX(ethylene-induced xylanase)和非生物逆境因子NaCl诱导下显著差异表达。对部分响应NaCl诱导的差异表达基因进行了qRT-PCR验证,结果暗示这些基因可能在响应逆境胁迫中发挥调控功能。这些结果将为进一步解析番茄RLCK的生物学功能提供参考信息。Receptor-like cytoplasmic kinases(RLCKs)are a class of receptor-like kinases that lack both transmembrane and extracellular domains and contain intracellular kinase domains.RLCKs play crucial roles in physiological process such as plant growth,development and defense responses.However,only a few tomato RLCKs have been characterized so far.In order to explore the evolution and role of the tomato(Solanum lycopersicum)RLCK family,121 RLCK genes were identified in the tomato genome and further classified into 17 subfamilies based on the phylogenetic analysis of the protein sequences.Bioinformatic analysis showed that tomato RLCK family members are diverse and conserved at the DNA and protein levels,respectively.Tomato RLCKs are unevenly located on 12 chromosomes,and fragment duplication drove the expansion of the tomato RLCK gene family.Tissue expression analysis indicated that tomato RLCKs exhibit different expression patterns at the stages of tomato growth and development,and tomato RLCKs show expression in all examined developmental stages.Microarray data analysis showed that a small number of tomato RLCKs were differentially expressed under biotic factor EIX(a fungal elicitor ethylene-induced xylanase)and abiotic factor NaCl treatments,respectively.The expressions of few differentially expressed tomato RLCKs in response to NaCl treatment were verified by qRT-PCR,suggesting that these genes may involve in tomato plant responses to stress.These data can lay a foundation for further investigation of the biological functions of the RLCK family members in tomato.
